中文摘要 | 臺灣2009年乳癌死亡率佔癌症死亡率的8%,排名女性癌症死亡率的第4位。分次輻射治療是癌症治療最常使用的治療手段,此研究結果旨在顯示輻射處理後細胞的存活率、細胞週期的分佈,及其分子傳遞訊號蛋白的表現,包括Chk2、硫胱氨酸蛋白酶-3(caspase-3)、基質金屬蛋白酶-2(matrix metalloproteinase-2)及NF-κB等。輻射暴露經2, 4 或6 Gy後之細胞存活率依序為84.1%, 80.2% 及 78.4%。結論顯示經輻射後MCF-7乳癌細胞所造成的G2/M期的停滯性乃藉由Chk2的表現所誘導形成。而細胞分裂死亡途徑可能藉由caspase-3增加媒介所造成。在2 ~6Gy輻射暴露後NF-κB蛋白及基質金屬蛋白酶表現比控制組還低且隨輻射劑量增加而下降,顯示此分次劑量下有利於臨床分次輻射治療可避免癌細胞之竄逃。 |
英文摘要 | Breast cancer was the fourth cause of cancer death in women and got 8% of cancer death in Taiwan 2009. Fractionated radiotherapy is a common treatment method for breast cancer therapy. In this study, results showed the radiation effects on viability, cell cycle distribution and related signal protein expression including Chk2, Caspase-3, matrix metalloproteinase-2 and NF-κB in human breast cancer cells. Viability of cancer cells treated by 2, 4 or 6 Gy irradiation got 84.1, 80.2 and 78.4% respectively. After MCF-7 cell exposure to radiation, it only caused G2/M arrest mediated from Chk2 expression and mitotic death passed through increases of caspase-3 expression. NF-κB expression after radiation was decreasing and dependent on increasing dose exposure. NF-κB expression after 2Gy irradiation decreased more than control group. It is good for clinical fractionated radiotherapy to prevented tumor cell escape. In conclusion, these results demonstrated that irradiation led to decrease the levels of matrix metalloproteinase-2 and NF-κB expression than control group to inhibit the cell migration. |
