中文摘要 | 背景:蜆(Corbicula fluminea)在亞洲地區為廣泛使用的食品,本草綱目中認為具有保護肝臟的作用。有關蜆萃取物在養生保健之功效近年來被廣泛的討論,然而相關的作用機轉仍未被清楚的探討。目的:探討蜆萃取物是否經由調節氧化壓力達到保肝的功效。方法:以清醒鼠失血性休克(Hemorrhagic shock;HS)動物實驗模式探討蜆萃取物對於失血引起的肝臟損傷之保護功效。研究設計將實驗動物隨機分成三組:HS+FC組:為失血40%後給與食用蜆萃取物20mg/kg;HS組:為單純失血組;第三組為控制組。以血清生化值AST、ALT、LDH評估肝臟損傷程度;以血清腫瘤壞死因子(tumor necrosis factor-alpha;TNF-α)評估發炎前趨物質的濃度;檢測血清氧自由基(free oxygen radicals test;FORT)與抗氧化力(free oxygen radicals defense;FORD)評估其調節氧化壓力功效。結果:蜆萃取物能降低血清TNF-α、AST、ALT與LDH濃度,對肝臟產生保護的功效(p<.05),而對於氧化壓力調節則無明顯的成效。結論:蜆萃取物可以經由減少發炎前趨物質血清TNF-α的產生降低肝損傷,然而氧化壓力調節效果則須要在進一步的研究確認。 |
英文摘要 | Background: The freshwater clam (Corbicula fluminea) is said to have beneficial effects on liver function. Recent reports indicate that freshwater clam extract (FC) has many bioactivities. However, investigations of FC involving in oxidative stress-mediated hepatic injury are limited.Purposes: The objective of this study was to assess the possible activity of FC in acute liver injury induced by hemorrhage in rats.Methods: The authors randomly divided male Wistar-Kyoto rats into three groups. Hemorrhagic shock (HS) was induced in the HS+FC (n=8) and HS (n=8) groups. The rats in the HS+FC group were fed 20 mg/kg FC. HS was not induced in rats (n=8) in the control group and they were not fed FC. Blood levels of white blood cells (WBC), aspartate aminotransferase (AST), alanine aminotransferase (ALT), lactic dehydrogenase (LDH), tumor necrosis factor-alpha (TNF-α), reactive oxygen species concentration (as free oxygen radicals test; FORT units) and FC antioxidant activity (as free oxygen radicals defense; FORD units) were measured at several time points during the experiment.Results: HS significantly decreased the mean arterial pressure, and increased blood AST, ALT, LDH and FORT levels and induced liver injury in rats. Treatment with FC extract decreased AST, ALT, LDH, and TNF-α levels after hemorrhage. However, no significance difference in the FORD concentration was found between the HS and HS+FC groups.Conclusions: FC has the ability to suppress the HS-induced release of pro-inflammatory cytokines TNF-α and decrease the serum levels of ALT, AST and LDH. However it remains unclear whether FC has an antioxidant effect. The antioxidant activity of FC in different regions of the world warrants further exploration. |
