中文摘要 | 本試驗目的在探討添加維生素C對受過氧化氫處理之H9c2(2-1)心肌肌母細胞所產生抗氧化作用之效果。由細胞存活率結果顯示,分別於12、24及48 h添加不同濃度維生素C(0、25、50、100、200μM)於H9c2(2-1)心肌細胞,隨著時間及濃度增加,細胞存活率並無顯著差異(p > 0.05),顯示在此範圍濃度下維生素C 並不具細胞毒性。以H2O2 處理H9c2(2-1)心肌細胞,其細胞存活率則隨著時間(1、3、6 小時)及濃度(0、100、150、200、300μM)之增加而顯著降低(P < 0.05)。以維生素C(0、25、50、100、200μΜ)前處理24 h後,再添加100μM H2O2作用3 h,結果顯示,添加100μΜ及200μΜ之維生素C有顯著抗細胞凋亡之能力(p < 0.05),細胞存活率分別可由30.5%提升至68.1%及71.3%。當添加H2O2(100μM)作用3 h,其細胞形態明顯皺縮;若以維生素C(100μM)前處理24 h後再添加H2O2(100μM)作用3 h,則明顯抑制細胞皺縮。在細胞內氧化壓力測定上,添加H2O2(100μM)與對照組比較,有顯著性上升(P < 0.05);以維生素C(100μΜ)前處理24 h後再添加H2O2(100μM)3 h,細胞內氧化壓力明顯由8.4倍下降至5.7倍(P < 0.05)。試驗結果證實,添加維生素C(100μM)對H9c2(2-1)心肌細胞於H2O2(100μM)處理下具有抗氧化之效果。 |
英文摘要 | The purpose of this study was to examine the antioxidant effect of vitamin C on hydrogen peroxide's (H2O2) stress in the cardiomyoblast cell line H9c2 (2-1). The results showed there was no harm to cell viability when the H9c2 (2-1) cells was treated with vitamin C (0, 25, 50, 100 or 200μM) for 12, 24 and 48 h cultivation. The percentage of cellular apoptosis of H9c2 (2-1) cells increased in a dose-and time-dependent manner when accompanied with H2O2 treatments (0, 100, 150, 200, 300μΜfor 1, 3 and 6 h). After pretreatment with vitamin C for 24 h and then with H2O2 (100μM) for 3 h, the cell viability of 100 and 200μM of vitamin C treatments were significantly higher than the untreated vitamin C group (p < 0.05). The cell viability rose from 30.5%to 68.1%and 71.3%, respectively. Significant cell shrinkage and oxidative stress in H9c2 (2- 1) cells were also observed in the treatment of 100μM H2O2 for 3 h (p < 0.05). The phenomenon of cell viability could be partially rescued by pretreatment with 100μM of vitamin C for 24 h before H2O2 (100μM, 3 h) was added. When pre-treated with vitamin C (100μΜ) for 24 h before adding H2O2 (100μM), intracellular oxidative stress decreased significantly from 8.4 times to 5.7 times (P < 0.05). In summary, the results revealed vitamin C could serve as an antioxidant for crdiomyoblast H9c2 (2-1) to overcome H2O2 challenges. |
