中文摘要 | 本研究主要調查中華絨螯蟹繁殖時之病害問題。1株造成種蟹大量死亡之革蘭氏陰性的溶血弧菌(Vibrio parahaemolyticus was)被由飼養於鹽度千分之15下的發病種蟹中分離出。此發病種蟹呈現厭食、活力不佳、肝胰腺壞死、肌肉白濁壞死、黃棕色且不正常凝固之血淋巴,和大量死亡。此次調查共計分離6株弧菌,其中有一株經API 20E鑑定為溶藻弧菌V.alginolyticus,其他的皆為溶血弧菌。菌株ES05具有最強的毒性,進一步經16S rDNA定序分析,獲得一段長度1513 bp的序列(GenBank accession no.: JN595786),經與基因庫中之其他溶血弧菌的16S rDNA分析比對顯示相似性遠99.7%~99.8% (GenBank accessionnos.: NC_004603 and NC_004605)。半致死毒性試驗顯示該菌對中華絨螯蟹種蟹之96小時致死濃度為7.8 × 10^3 colony-forming units (g crab)^(-1)。鹽度緊迫分析發現,當中華絨螯蟹被置於鹽度千分之15下進行繁殖時,其對菌株ES05的感受性會明顯提高。這意昧著,中華絨螫蟹被置於半淡海水中促產繁殖時,如果同時感染弧菌,將引起更嚴重的死亡。 |
英文摘要 | A disease survey of Chinese mitten crab, Eriocheir sinensis, placed in saltwater (15 per mill) for mating and spawning in a reproductive period was conducted in this study. Sixteen isolates of Vibrio were isolated from diseased crabs which displayed anorexia, inactivity, erosion of the hepatopancreas, whitish muscle with erosion, abnormal coagulation of hemolymph with a yellowish-brown color, and mortality. One of the isolates was identified as V. alginolyticus, and the others were V. parahaemolyticus by the API 20E system. Strain ES05 with higher virulence was further confirmed by a 16S rDNA sequencing analysis, and its 16S rDNA sequence (1513 bp) (GenBank accession no.: JN595786) exhibited 99.7%~99.8% sequence identity to V. parahaemolyticus (GenBank accession nos.: NC_004603 and NC_004605). The calculated 96 h 50% lethal dose was 7.8 × 10^3 colony-forming units (g crab)^(-1). The test of disease susceptibility of crabs to V. parahaemolyticus ES05 showed that crabs under salinity stress (15 per mill) had significantly higher mortality than control crabs reared in fresh water. Thus, it is recognized that transfer of pathogen-carrying broodstock to saltwater for reproduction may result in a disease outbreak and consequently in a high mortality. |
