中文摘要 | 研究目的:口腔癌是一種預後很差的癌症,在過去二十年口腔癌的平均五年存活率並沒有顯著的改善.在腫瘤生成中,血管新生是一個相當重要的機轉,其中會影響的因子包含了血小板衍生性生長因子.血小板衍生性生長因子和細胞週期調控及細胞凋零有關.在目前的研究中顯示,血小板衍生性生長因子和其受體的基因表現和許多疾病及癌症的進程有關.本研究之目的就是探討血小板衍生性生長因子及其受體在口腔鱗狀細胞癌之基因表現.研究方法:本研究收集40位在台北馬偕紀念醫院接受手術治療的病人,切除之鱗狀細胞癌組織為實驗組以及周邊無腫瘤組織為對照組等40對組織,以定量反轉錄PCR方式分析人類血小板衍生性生長因子(PDGFA、PDGFB、PDGFC、PDGFD)及其受體(PDGFR-A及PDGFR-B)之基因表現,再以統計方法分析腫瘤組織和周邊無腫瘤組織基因表現程度之不同.結果: 血小板衍生性生長因(PDGFA、PDGFB、PDGFC)及其受體(PDGFR-B)之基因在口腔鱗狀細胞癌組織有顯著的過度表現(P<0.05).這樣的結果顯示出血小板衍生性生長因子(PDGFA、PDGFB、PDGFC)及其受體(PDGFR-B)在口腔鱗狀細胞癌發展生成上應扮演相當重要的角色.結論: 血小板衍生性生長因(PDGFA、PDGFB、PDGFC)及其受體(PDGFR-B)之基因在口腔鱗狀細胞癌組織有顯著的過度表現,在腫瘤形成上扮演相當重要的角色,其中PDGFC及PDGFR-B在癌症組織的過量表現是較為新穎的發現,PDGFR-B所引發的相關機轉值得進一步探討. |
英文摘要 | Objective: Oral cancer is a kind of cancer with poor prognosis. In Taiwan, oral cancer is the 6th highest among all cancers in causes of death, and the mortality rate is the 4th highest in male. According to the statistics, the 5-year survival rate haven't been improved significantly during past 20 years. Angiogenesis is one of the most important mechanism in tumorogenesis. Platelet-derived growth factor (PDGF) is one of the factor that would induce angiogenesis. PDGF is also involved in cell cycle adjustment and apoptosis. Our aim is to analyze the PDGF and PDGFR genes expression in oral squamous cell carcinoma and relative carcinogenic factors. Material & Methods: We collected 40 pairs of oral squamous cell carcinoma tissue as experimental group and non-cancerous matched tissue as control group from 40 patients who received surgical excision treatment in Taipei Mackay Memorial Hospital. We analyzed the PDGFA,PDGFB, PDGFC, PDGFD, PDGFR-u, and PDGFR-p genes expression with qRT-PCR and compare the results from cancer tissue and non-cancerous tissue. Results: Our result showed significant over-expression of PDGFA, PDGFB, PDGFC, and PDGFR-p genes expression between the squamous cell carcinoma tissues and the noncancerous matched tissues (p < 0.05). PDGFA,PDGFB, PDGFC, and PDGFR-p may play an important role in oral squamous cell carcinoma tumorigenesis. Conclusion: PDGF A, PDGFB, PDGFC, and PDGFR-p genes are significantly over-expressed in oral squamous cell carcinoma tissue. PDGFC and PDGFR-p genes over-expression are novel result in oral cancer research. But we still need further work on the relative mechanism induced by PDGFR-p. |
