中文摘要 | 本試驗之目的乃研究豬卵母細胞於體外成熟培養時間,對豬卵母細胞於體外成熟、核移置重組卵之激活暨隨後胚發育之影響。豬卵丘-卵母細胞複合體取自於豬卵巢,並培養於NCSU-23成熟培養液中0~60小時。新鮮卵丘細胞取自於卵丘-卵母細胞複合體,並培養於含有10%胎牛血清(fetal calf serum, FCS)之DMEM培養液中,繼代培養至第4代(F4)以作為供核體細胞之來源,注入於去核卵母細胞之卵質內邊緣以獲得重組卵。核移置後,重組卵同時進行融合與激活,隨後並培養於NCSU-23胚培養液中16小時觀察重組卵激活情形或培養144小時觀察胚發育情形。結果發現,卵母細胞之成熟培養時間顯著影響卵母細胞成熟率、與重組卵之激活率、原核形成率與胚發育,48小時為卵母細胞最佳之成熟培養時間。 |
英文摘要 | The effect of in vitro maturation time of porcine oocytes on the activation and development of reconstructed porcine oocytes with cultured cumulus cells as donor cells was studied. The cumulus-oocyte complexes (COCs) obtained from porcine ovaries were cultured in NCSU-23 maturation medium for 0-60 h. The fresh cumulus cells separated from COCs were cultured in DMEM medium (10% fetal calf serum, FCS) for up to 4 passages. The cultured cumulus cells were injected into the enucleated oocytes to obtain the reconstructed oocytes. After nuclear transfer, the reconstructed oocytes were fused and activated simultaneously. Then, the activated oocytes were cultured in NCSU-23 embryo culture medium for 16 h to observe the activation or 144 h to observe the embryo development. It was found that the maturation time affected the activation rate, pronuclear formation rate and embryo development rate of the reconstructed oocytes significantly. A maturation time of 48 h was found to be an optimum condition. |
