中文摘要 | 本試驗之目的,乃研究不同濃度之乙醇(0、3、5、7與9%)與選定濃度之乙醇(7%)以不同時間(0、1、3、5與7分鐘)測試對體外成熟豬卵母細胞激活之影響。豬卵母細胞經體外成熟培養48小時後,分別置於不同濃度之乙醇各5分鐘與置於7%乙醇中各0、1、3、5 與7分鐘,進行豬卵母細胞之激活刺激,隨後並培於NCSU-23培養液養分別培養16小時與144小時觀察卵母細胞激活與胚發育情形。試驗結果呈現,7%乙醇分別以0、1、3、5 與7分鐘對豬卵母細胞激活之卵分裂率分別為7.6、12.0、44.9、63.0與52.3%,以5分鐘組之胚發育比率最高(P < 0.05),胚囊發育比率也以5分鐘組最高(P < 0.05),顯示體外成熟之豬卵母細胞可以僅用7%乙醇施以激活5分鐘的最佳處理條件,獲致顯著的體外胚囊發育進展。 |
英文摘要 | The objective of this study was to investigate the activation effects at various concentrations of ethanol (0, 3, 5, 7 and 9%) for regular exposure duration (5 minutes) and as well at specified concentration of ethanol (7%) for various exposure duration (0, 1, 3, 5 and 7 minutes) on the in vitro matured porcine oocytes. Porcine oocytes separated from same preparation batch were exposed for 5 minutes at various ethanol concentrations and as well were exposed for 0, 1, 3, 5 and 7 minutes at 7% ethanol after maturation in culture for 48 hours. Subsequently, both activation and embryo development of the activated oocytes were respectively observed for 16 h and for 144 h in NCSU-23 embryo culture medium. The results indicated that the cleavage rates upon oocytes activation for 0、1、3、5 and 7 minutes group by 7% ethanol were 7.6、12.0、44.9、63.0 and 52.3%, respectively. Moreover, the observed highest cleavage rates upon 5 minutes exposure among other groups (P < 0.05) has also shown higher blastocyst rates among other groups (P < 0.05). The present study indicated that the 7% ethanol only exposure for 5 minutes may serve optimal activation on in vitro matured porcine oocytes with significant in vitro blastocyst progression. |
