英文摘要 | The proposed flow-through biosensing system was based on the competitive inhibitory activity of the analytes against L-amino acid oxidase (EC 1.4.3.2; from the venom of eastern diamondback rattlesnake Crotalus adamanteus). The enzyme was covalently immobilized on porous glass beads with a high surface/volume ratio (80-120 mesh, 70 nm pore diameter); the enzyme glass beads were then packed as a mini-reactor (2 mm × 5 cm) suitable for flow-injection (FIA) or liquid chromatography (LC) system. The mini-bioreactor was used to construct an amperometric FIA system; the dynamic range of the analytical system for benzoate determination was from tens of μM to mM order with CV < 3%. The system sensed also several physiologically unusual chemicals of medical interest, and the mini-bioreactor was durable for at least 12 months. |
