英文摘要 | Barringtonia asiatica (L.) Kurz. is a seashore plant with fruits which usually drift on the sea. This species is widely distributed in the palaeotropic and northward extending to Hengchun Peninsula and Lanyu. However, due to overharvest of individual trees for wood and habitat destruction, the populations of this species in Taiwan and Lanyu have gradually declined. In this study, the genetic variation and population structure of 69 individuals sampled from seven populations of B. asiatica in Hengchun Peninsula and Lanyu were studied using ISSR markers. POPGENE analysis revealed total gene diversity (H) was 0.2041 and the genetic differentiation index (Gst) was 0.3870, while the gene flow index (Nm) was 0.3959. Analysis of molecular variance (AMOVA) revealed the variance component between population regions was 13.03% (P<0.05(superscript *)), the populations within regions was 26.17% (P<0.001(superscript **)) and among individuals within the population it was 60.81 (P<0.001(superscript **)). The result indicated significant genetic differentiation between Hengchun Peninsula and Lanyu and among population within regions. However, the greatest variation was among individuals within populations. UPGMA cluster analysis and principal coordinates analysis (PCoA) showed they could be divided into two groups: Taiwan group and Lanyu group. In addition, there was no significant relation between genetic distance and geographic distance (r=0.44, p=0.98), pointing to a random distribution of individuals among populations. In conclusion, the gene flow among populations was relatively restricted and the populations in different islands showed a significant genetic differentiation. Based on the result of UPGMA cluster analysis for the population cluster within the island, geographic segmentation may be a significant factor in the genetic differentiation between the two regions. The low genetic diversity indicated species between Hengchun Peninsula and Lanyu had suffered the bottleneck effect. Based on the above finding, both the In-situ and Ex-situ conservation approaches are necessary in preserving this species. |
