中文摘要 | 肝醣合成酶激酶-3(Glycogen synthase kinase 3; GSK-3)是存在於所有的真核細胞中的一種serine/theronine激酶(Kinase)。此酵素不但能夠促使肝醣合成酶(glycogen synthase)磷酸化及抑制肝醣的合成,並且可以藉由磷酸化調控許多重要蛋白質的功能。本文回顧近年來的文獻,闡述GSK-3如何影響訊息傳導的途徑,胚胎的分化,細胞的增生,細胞凋亡,及癌化。此外,也一併討論GSK-3基因缺失的影響,GSK-3a and GSK-3b在功能上可能的差異,以及目前以GSK-3為標的,在發展藥物上的進展。本篇文章將有助於對GSK-3的重要性有更進一步的了解。 |
英文摘要 | Glycogen synthase kinase 3 (GSK-3) is a serine/threonine kinase that is present in all eukaryotic cells. This enzyme phosphorylates not only glycogen synthase to suppress enzyme activity and glycogen synthesis, but also a wide range of proteins in cells, modulating their functions. This article elucidates the mechanism by which phosphorylation by GSK-3 influences signaling pathways, embryogenesis, cell proliferation, apoptosis, oncogenesis, and the functions of transcription factors. The effects of genetic defects on the GSK-3 gene, functional differences between GSK-3 and GSK-3 and the recent advances on drug development, using GSK-3 as a drug target, are discussed. This article offers useful information about the importance of GSK-3. |
