中文摘要 | 本研究之目的係以阿里山十大功勞(Mahonia oiwakensis)及台灣黃蘗(Phellodendron amurense)兩種台灣特有種之傳統藥用植物的根、莖與葉為試材,經冷浸法或熱迴流萃取法取得其乙醇萃取液,以探討其對常見病原菌類(Staphylococcus aureus, Bacillus subtilis, Escherichia coli)等之抑菌性, 並評估其於溫度與酸鹼變化之抑菌效果。另利用高效能液相層析儀(High Performance Liquid Chromatography, HPLC)分析各萃取物之小蘗鹼(Berberine)含量。結果得知,兩試材之各乙醇萃取液對Staphylococcus aureaus BCRC 10451、Bacillus subtilis BCRC 10255及Escherichia coli BCRC 10675等均有抑菌性,且對Staphylococcus aureaus BCRC 10451之抑菌效果最佳。除阿里山十大功勞葉部之熱迴流萃取液外,所有乙醇萃取液於0.1mg/mL濃度時,對 Staphylococcus aureaus BCRC 10451具有抑制效果,其抑菌圈直徑介於 11.98-15.01mm。另於耐熱性試驗結果顯示,經溫度變化之冷浸法所得乙醇萃取液對Staphylococcusaureaus BCRC10451仍具有抑菌效果,故其主要抑菌成分具有良好之熱穩定性。而在酸鹼環境變化對抑菌性影響之結果顯示, pH值於5-11的環境下, 兩試材根部以冷浸法所得乙醇萃取液對Staphylococcus aureaus BCRC10451具有抑菌效果,且其隨pH值提高而增加。HPLC分析結果得知,以台灣黃蘗根部於熱迴流萃取者之小蘗鹼含量417.34 mg/g為最高;而阿里山十大功勞葉部者之含量17.32 mg/g最低。 |
英文摘要 | The purpose of this study was to investigate two types of plants used in traditional Chinese medicine in Taiwan, Mahonia oiwakensis and Phellodendron amurense. The experimental materials included the root, stem, and leaf of the plants. The extracts were obtained by using the cold soaking process and the hot backset refined method with ethanol. The extracts were used to investigate their antibacterial (Staphylococcus aureus, Bacillus subtilis, Escherichia coli) activity. The effects of temperature and pH value on their antibacterial activities were also examined. The content of berberine from these ethanolic extracts was analyzed by using high performance liquid chromatography (HPLC). The experimental results of the antibacterial tests showed that the extracts were all effective for antibacterial activity of Staphylococcus aureus BCRC 10451, Bacillus subtilis BCRC 10255, and Escherichia coli BCRC 10675. The best antibacterial activity was for Staphylococcus aureus BCRC 10451. The minimum inhibitory concentration for all extracts was 0.1mg/mL, effective for all bacteria used. The diameter of growth inhibition was between 11.98-15.01 mm by using the hot backset refined method with ethanol, except for the extracts from the root of Mahonia oiwakensis. The extracts, using the cold soaking method with ethanol from the leaf of Mahonia oiwakensis, were tested with Staphylococcus aureus BCRC10451, after the temperature changed. The results showed that the antibacterial activity was effective, and was not influenced by the temperature change of the extracts. The effect of pH value on the antibacterial activity showed that the extracts of the root, using the cold soaking method, from both experimental materials inhibited Staphylococcus aureus BCRC 10451. In addition, the antibacterial activity increased with increasing pH value from 5 to 11. The results of the HPLC analysis showed that by using the hot backset refined method, the highest content of berberine was obtained for the extracts from the root of Phellodendron amurense, about 417.34 mg/g, and the extracts from the leaf of Mahonia oiwakensis was only about 17.32 mg/g. |
