中文摘要 | 本研究選取台灣常見之食用鳳梨(Ananas comosus L. Merrill)台農20號之腋芽為材料,利用已建立之高效率癒合組織誘導與植株再生培養系統,進行農桿菌介導基因轉殖系統之建立。初步試驗結果顯示,將腋芽培養於同時添加2 ppm BA與NAA之MS 基礎培養基中可誘發擬球體(protocorm-like body, PLB)癒合組織,每個月繼代培養以獲得足夠數量之PLB,再進行基因轉殖。另外,試驗結果顯示利用超音波震盪輔助可大幅提高農桿菌轉殖效率至30%,顯示超音波震盪有助於改善部分較難建立之作物轉殖系統,甚值得深入研究。 |
英文摘要 | The axillary buds from Taiwanese edible pineapple (Ananas comosus L. Merrill cv. Tainung 20) were used in this study. The buds were inoculated on MS basal medium supplement with 2 ppm of BA and 2 ppm of NAA (MSB2N2) for protocorm-like body (PLB) induction. The callus were subculture every month at the same medium for PLB propagation. At present study, we used sonication-assisted Agrobacterium-mediated transformation (SAAT) method to enhance the transformation efficiency. The transformation ratio is approximately to 30% by transit GUS staining analysis and PCR analysis. The SAAT method would be useful to improve the transformation efficiency on some crops. According to these result, we develop an effective shoot regeneration and transformation system in Taiwanese edible pineapples. Further research is undergoing on molecular farming and edible vaccine production. |
