英文摘要 | It is well established that coculture with Vero cells improves embryo development. Several studies suggest that LIF (leukemia inhibitory factor), PDGF (platelet derived growth factor), TGF (transforming growth factor) α and β (now known as TGFβ1), and EGF (epidermal growth factor) play an important role in embryo development. In order to quantify the cytokine and growth factors produced by Verocell coculture, Vero cells were seeded into 6 wells in NUNC 4-well dishes in different culture mediaplus 5% fetal bovine serum. Culture medium was collected from each well every day for 6 days. Unconditioned medium without Vero cells was used as a control. The culture media used included human tubal fluid (HTF) medium, Menezo's B2 medium, and Medi-cult universal IVF medium. With Vero cell cocultures, LIF was elevated from day 1 but remained almost constant during the culture interval; PDGF increased rapidly from day 1 through day 6. TGF β1 increased from day 1 to day 3, but plateaued thereafter. EGF, however, was almost undetectable, Unconditioned medium produced little of these cytokine and growth factors. We speculate that these cytokines and growth factors play a role in the embryotrophic action of Vero cell coculture. |
