中文摘要 | 本文是在水溫24℃下,以Oxolinic acid對點帶石斑(Epinephelus coioides) 進行單次口投給藥的吸收、分佈與排除之研究,口投劑量為40 mg/kg魚體重。Oxolinic acid之含量以高效能液相層析儀檢測,並以一室式動力學模式解析藥物在魚體內的吸收、分 佈與排除情形。結果發現,Oxolinic acid在魚體內各組織器官的吸收皆在12 h內達最高 峰,其最高含量的多寡為腎臟>肝臟>血液>肌肉。另外,Oxolinic acid在血液中達高峰 的濃度及時間分別為1.45μg/ml及5.75 h,半衰期為15.1 h,血液的曲線下面積和可用率分 別為37.1μg.h/ml及18.1%,而Oxolinic acid在血液、肌血、肝臟及腎臟內的預估排除期 (檢測下限為0.001μg/ml)則分別為162、200、200及254 h,亦即以上述條件處理點 帶石斑,停藥期應為11天。 |
英文摘要 | The absorption, distribution, and elimination of oxolinic acid were studied on orange-spotted grouper, Epinephelus coioides, after single oral administration at a dose of 40mg/kg in seawater at 24℃. Oxolinic acid levels were determined by a high performance liquid chromatographic method with the detection limit of 0.001μg/ml. Serum levels of oxolinic acid were described using a one-compartment pharmacokinetics model. The theoretical time to peak serum concentration of drug, Tmax, was 5.75 h and peak concentration, Cmax, at Tmax was 1.45μg/ml. The elimination half-life, t1/2, was 15.1h. The area under the serum concentration-time curve, AUC, and the bioavailability, F, were 37.1 μg.h/ml and 18.1%, respectively. Drug absorption was complete in about 12 h following dosing and the highest concentrations were measured in the kidney followed by the liver, muscle and serum. The predicted withdrawal period, Tpred, for oxolinic acid from serum, muscle, liver and kidney were 162,200,200 and 254 h, respectively. Based on the results of this study, the best withdrawal period is, therefore, 11 day. |
