中文摘要 | 飼料配方中使用多種穀類原料,黃麴毒素普遍發生於穀類原料,文獻記載以黃麴毒素B₁ (Aflatoxin B₁, AFB₁))為這類黴菌毒素毒性最強,因此,對於飼料中黃麴毒素含量有瞭解的必要。本試驗中使用免疫吸附管柱(immunoaffinity column)純化並搭配液相層析串聯式質譜儀(Liquid chromatography/tandem mass spectrometry, LC/MS/MS),發展出具有靈敏度及選擇性之黃麴毒素分析方法,使用三段式四極棒質譜儀(Triple quadruple mass spectrometry)搭配渦輪噴灑離子源(TurboIon Spray source)在正離子的模式下偵測黃麴毒素。整體分析回收率分別為:黃麴毒素B₁ 43.52%、黃麴毒B₂ (Aflatoxins B₂, AFB₂,) 80.70%、黃麴毒素G₁ (Aflatoxins G₁,AFG₁) 81.62%以及黃麴毒素G₂ (Aflatoxins G₂, AFG₂) 87.28%,其變異係數(coefficient of variation, CV),範圍介於6.43%至9.20%之間。四種黃麴毒素之偵測極限(LOD, S/N = 3)為0.06~0.11 ng/mL,四種黃麴毒素之定量極限(LOQ, S/N = 10)為0.25~0.69 ng/mL,儀器對於四種黃麴毒素之CV介於3.33%至5.63%之間。本方法可應用於飼料化驗分析。 |
英文摘要 | Most feeds are formulated with cereals and grains. Aflatoxins occur widely in cereals and grains. Aflatoxin B₁ (AFB₁) is well known as the most toxic of this class of mycotoxins. So, it is necessary to determine the amount of aflatoxins in feeds. In this experiment, we coupled the method of immunoaffinity column clean-up with liquid chromatography/tandem mass spectrometry (LC/MS/MS) to produce a sensitive ans selective approach for the analysis of aflatoxins. A triple-quadruple mass spectrometry using a TurboIon Spray source operating in the positive ion mode was used to detect aflatoxins. Recoveries of the full analytical procedure were 73.52% for aflatoxins B₁, 80.70% for aflatoxins B₂, 81.62% for aflatoxins G₁ and 87.28% for aflatoxins G₂, respectively, with the coefficient of variation (CV) values ranging from 6.43% to 9.20%, The limits of detection (LOD, S/N = 3) ranged from 0.06 to 0.11 ng/mL for the four aflatoxins. The limits of quantification (LOQ, S/N = 10) ranged from 0.25 to 0.69 ng/mL for the four aflatoxins. The CV values for the four aflatoxins were from 3.33% to 5.63%. We concluded therefore that this method can be applied to the analysis of aflatoxins in feeds. |
