| 中文摘要 | 94 年度引進即時核酸偵測儀(Real-time PCR),並建立即時定量聚合酶鏈反應診斷技術,初步應用於犬小病毒之核酸偵測,模板DNA用量為100 ng、10ng、1 ng、100 pg、10 pg、1 pg皆有螢光訊號,最低可測得至1 pg之核酸濃度,而核酸與循環數之標準曲線,可應用於核酸定量;另已收集美國印第安那州動物疾病診斷實驗室(Indiana animal disease diagnostic laboratory)及肯塔基州家畜疾病診斷中心(Kentucky livestock disease diagnostic center)的實驗室資訊管理系統相關資料,如檢體送檢流程、檢驗標準流程與系統操作介面等,可資往後本所規劃動物防檢疫自動化資訊系統之參考。 |
| 英文摘要 | We have set up the real-time PCR machine and the procedure of the nucleic acid detection with real-time polymerase chain reaction (PCR). The procedure was applied to detect canine parvovirus firstly. The level of nucleic acid which could be detected by the constructed assay in this study was as low as 1 pg. The standard curve about the relationship between concentration of nucleic acid and cycle numbers could be applied to quantify the nucleic acid. Information provided from Indiana Animal Disease Diagnostic Laboratory (ADDL) and Kentucky Livestock Disease Diagnostic Center (LDDC) can be applied successfully to set up an automated information system for animal inspection and quarantine in the future. |
