中文摘要 | 本研究的目的係藉由動物實驗,探討對誘導產生的糖尿病Wistar鼠藉由鉻酵母的補充,是否會影響其血糖及血脂狀況。雄性Wistar鼠40隻,隨機分為4組,分別為NA:AIN-76飲食控制組、NC:銘飲食控制組、DA:AIN-76飲食糖尿病組及DC:銘飲食糖尿病組。在進入實驗期前,先測Wistar鼠之空腹血糖、果糖胺、三酸甘油酯、總膽固醇、高及低密度脂蛋白,以建立基礎值。實驗期為28天,同時將鉻酵母利用管灌方式投與補充鉻組,每日早晨十時投與每隻Wistar鼠600mg/kg BW的三價鉻。第2型糖尿病Wistar鼠的誘導是以腹腔注射兩劑 (連續兩天) STZ 65 mg/kg BW,在注射前的分鐘先注射nicotinamide (230 mg/kg BW),待一星期後,測空腹血糖值,數值大於180mg/dL認定有糖尿病。四週後,取血測血液上述參數,並進行比較。結果發現,每日補充600mg/kgBW的鉻酵母,時間為期四週,對於由STZ 誘導產生的糖尿病Wistar鼠,並未觀察到有改善血糖及血脂的狀況,其原因可能是與所使用之動物模式、投與鉻酵母的的劑量及時間有關,但詳細機制尚須更進一步的研究。 |
英文摘要 | The aim of this study was to evaluate the effect of a chromium yeast diet on blood glucose and lipid in streptozotocin (STZ)-induced diabetic Wistar rats. Forty male Wistar rats were randomly divided into four groups; they were NA (AIN-76 diet control), NC (chromium diet control), DA (AIN-76 diet DM) and DC (chromium diet DM). Before the experiment, they should be measured the fasting blood glucose, fructosamine, triglyceride, total cholesterol, high and low density lipoprotein to be the baseline. Experimental period were 28 days. We treated the trivalent chromium 600μg/kg BW by gavage in chromium diet groups at 10 o’clock on everyday. The DM groups were injected with nicotinamide (230 mg/kg BW) and STZ (65 mg/kg BW) to induce diabetes. We measured the fasting blood glucose after one week and diabetes mellitus was identified when the fasting blood glucose value was higher than 180 mg/dL. After 4 weeks of feeding, a significantly change of STZ-diabetic rats both with and without chromium yeast were hyperglycemic. But no significant changes in glucose metabolism and lipid parameters were observed. The reason may include the animal mode and the dose and the time of chromium yeast supplement, however, the detail mechanism needs further study. |
