中文摘要 | 以‘Blush Hawaii’菊花及其他品種試管苗為材料,將葉片橫切培養,觀察培植體不定芽的形成,並將所產生不定芽經發根馴化後移至田間,觀察產生變異之現象。不定芽的再生以 MS 為基本培養基,添加 TDZ 及 Kinetin,TDZ 及 2iP,TDZ 和 BA。大部份處理均可誘導產生不定芽,其再生比率為 0-26.3%;而平均每個培植體產生的不定芽數為 0-19.5 個。以 MS + 0.5 mg/l TDZ + 1 mg/l 2iP 培養基比較 17 個品種的再生,其中‘橙精興之華’、‘黑咖啡’、‘阿萊粉’、‘荷蘭白’、‘寒紅梅’、‘Regal Westland’、‘Cocarde’、‘Tiger’、‘Paso Doble’、‘Improved Mundial’、‘Salmon Impala’、‘Fiji’等 12 個品種均可產生不定芽。以‘寒紅梅’所產生的不定芽數最多,每個培植體平均產生 8.5 個不定芽,再生比率為 20%;‘Fiji’品種次之,每個培植體平均產生 7.3 個不定芽,再生比率為 18.8%。 |
英文摘要 | The purpose of this study was to establish a regeneration protocol from in vitro larnina culture of chrysanthemum shoots. In vitro shoots of Blush Hawaii and other cultivars were used as starting materials for leaf explant source for adventitious shoot formation. The established plantlets were also grown in the field to evaluate somaclonal variation. Murashige and Skoog medium was used as basal medium supplemented with various concentrations of thidiazuron (TDZ), kinetin, 2-isopentenyl adenine (2iP) and benzyladenine (BA). Adventitious shoots were regenerated from most treatments ranging at 0-26.39% and average of 0-l9.5 shoots per explant. One medium, MS + 0.5 mg/l TDZ + 1 mg/l 2iP, was chosen for comparison of 17 varieties for regeneration response. Among tested cultivars, a total of 12 cultivars regenerated shoots, including Cheng-Jin-Hsin-Tze-Hua, Hei-Ka-Fei, Ah-Lai Pink, Holland White,Cold Homae, Regal Westland, Cocarde, Tiger, Paso Doble, Improved Mundial, Salmon Impala and Fiji. The cultivar Cold Homae was most responsive, with regeneration rate of 20% and average 8.5 shoots per explant. Fiji responded next, with regeneration rate of l8.8% and average 7.3 shoots per explant. |
