中文摘要 | 本研究利用HPLC, potodiode array檢測器及高速原子衝擊質譜儀(FAB-MS Spectrometry),分析鑑定綠藻中葉綠素及其酵素(chlorophyllase)水解衍生物之組成及其構造。 綠藻細胞經破碎後,在70℃保溫30分鐘,總共有8種成份,即chlorophyll a, chlorophyll b, chlorophyllide a, chlorophyllide b; pheophorbide a, pheophorbide b, pheophytin a和pheophytin b可被檢測出,經photodiode-array檢測器檢測其吸收光譜,並以FAB-MS,分析其質譜,再與菠菜之葉綠素及衍生物比較,發現二者主要葉綠素群之色素及其酵素水解之衍生物,其中主要成份並無差別。 以同法分析市售綠藻片,發現大部份產品,經適當殺菁及加工處理後,葉綠素仍為其主要成份(40~60%),少量為脫鎂葉綠素(20~40%),及微量之酵素水解產物。但少數水份活性高於0.5以上之綠藻片,則發現酵素水解產物如葉綠酸(0.44~1.76%),脫鎂葉綠酸鹽(2.97~10.53%),有些微增加之趨勢,且脫鎂葉綠素含量亦有增加之現象,顯示在貯藏不當之產品中,酵素有可能恢復部份活性,影響產品品質。 |
英文摘要 | The composition of the chlorophylls and their enzymatic hydrolyzed derivatives in incubated chlorella extract were characterized by using both HPLC equipped with photodiode array detector and FAB Mass spectrometry. Chlorophyll a and b, chlorophyllide a and b, pheophorbide a and b, pheophytin a and b, were tentatively characterized by comparing the absorption spectrum between that of chlorella and spinach. The structure of the chlorophylls and their derivatives were confirmed by the melecular ion and some major fragments in the FAB Mass spectra. Most commercial available chlorella tablets contains mainly chlorophylls (40%~60%), and with minor amount of pheophytin (40~20%) and trace amount of pheophorbides. In some unusual samples, with a water activity higher than 0.5, significant increase in emzymatic hydrolyzed derivatives, pheophorbide (2.97~10.53%) and chlorophyllide (0.44~1.76%) were observed. These deta indicated that activity of chlorphyllase may be slightly activated under some storage condition for chlorella tables. |
