中文摘要 | 本研究利用自東港水試所分離之本土性等鞭金藻 Isochrysis galbana TK2,以 一次只改變基本培養條件中一因子之方式,獨立探討個別因子對其細胞生長、脂質生產及二 十二碳六烯酸產量之影響。結果發現隨培養時間增加,二十二碳六烯酸產量漸增至第八天止 ;最適培養溫度為 25 ℃; 最適光照條件為以 10,000lux 光強度進行連續光照;最佳氮源 為尿素,其最適濃度為 0.5mM;磷源 (KH �� PO �� ) 之最適濃度為 0.25g/L;螯合劑 (Na �� EDTA) 之最適濃度為 0.05g/L。 將上述各種最適因子組合之後,培養八天可得藻體乾重 為 0.52g/L,二十二碳六烯酸佔總脂肪酸之含量為 13.50%,產量高達 6.95mg/L。 |
英文摘要 | This study investigated the effects of cultivation conditions on the cell growth, lipid production and docosahexaenoic acid (DHA) yield of Isochrysis galbana TK2, a domestic marine microalga isolated by the Tungkang Kung Marine Laboratory. Each variable of interest was examined independently at different levels by holding the other variables constant to a basal set of conditions. The results showed that: the DHA yield increased with cultivation time until the 8th day, optimum cultivation temperature was 25 ℃, optimum lighting was achieved by continuous illumination at 10,000lux, urea was the best nitrogen source with a choice concentration of 0.5mM, the ideal concentration of phosphorous source (KH �� PO �� ) was 0.25g/L, and the best concentration of chelating agen (Na �� EDTA) was 0.05g/L. These individually identified optima of each variable were combined in the final study, and the cell mass, the DHA content of the lipid and the DHA yield after 8 days reached 0.52g/L, 13.5% and 6.95mg/L, respectively. |
