中文摘要 | 本試驗的目的為以RAPD技術分析4個青?種(系)間之遺傳親緣關係,建立區分青?品種之分子標誌,並探討品種親緣性與耐熱性之關係。從200組10 mer核?酸的逢機引子中篩選出13組(2.3%)引子(OPAB-2,OPAB-4,OPAB-10,OPAB-12,OPAB-13,OPAB-18,OPAB-19,OPAB-20,OPG-2,OPAR-10,OPAQ-11,OPG-13,OPG-19),可在4種青?品種(系)之間產生具有多型性的DNA條帶,所產生的RAPD圖譜可以清晰的將供試的4品種加以區分。13組引子共可產生83個DNA條帶,其中43個具有多型性。其中3組引子可區別4個品種,6組引子可區別3個品種,4組引子可區別2個品種。OPAB-19-180、OPAQ-11-920、OPAQ-11-650條帶為耐熱品種(`新莊`與`福?`)特有的條帶。連鎖群分析的結果顯示4種青?品種(系)之相似性係數介於0.78與0.61之間。以UPGMA程式進行相似性的群體分析,所得的結果顯示,四個青?品種具有高遺傳相似性,可概分為三群組:`蘭陽1號`與`宜蘭2號`屬於第一群組,`福?`為第二群組,`新莊`為第三群組。試驗結果顯示RAPD的分子標誌可以有效的區分青?4個品種(系),且其耐熱特性亦可部份反應在遺傳親緣關係。 |
英文摘要 | The purposes of this study were to determine the genetic relationship among four green onion (Allium fistulosum L.) cultivars, and to reveal the association between their genetic background and heat-tolerance by RAPD markers. Out of 200 decamer nucleotide primers, 13 primers showed consistent banding patterns and amplifications, that produced 83 bands with 43 polymorphisms. Four, three and two cultivars were discriminated by 3, 6, 4 primers, respectively. Three bands, OPAB-19-180, OPAQ-11-920, OPAQ-11-650, were only found in the heat-tolerant cultivars (‘Fu-tsun’ and ‘Hsinchuang’). The obtained among 4 green onion cultivavs similarity coefficients were between 0.61 and 0.78. The dendrogram revealed that four green onion cultivars can be classified into three groups, the first group includes ‘Lanyang #1’ and ‘Yilan #2’; the second group includes ‘Fu-tsun’; the third group includes ‘Hsinchuang’. This result supported that RAPD marker could precisely and effectively distinguish four green onion cultivars. The heat-tolerant characteristics could be partially associated with the genetic relationships. |
