中文摘要 | 本研究探討環境因子對培養紅麴菌Monascus ruber ATCC 18199生產其二次代謝物monacolin K(為一種膽固醇合成抑制劑成份)之影響。搖瓶實驗結果顯示,在前培養48小時後,將10%孢子懸浮液接種至生產培養基為最佳;本研究並發現菌株之monacolin K生合成是在紅麴色素生產之後四天;當生產培養基中之glycerol濃度增為176.4 mg/L時,可生產相對控制組2.7倍之monacolin K,且當glycerol對peptone之比值(即G/P ratio)為33時,monacolin K產量比控制組可增加為3.52倍,但當G/P比值達100可能因氮源受限而有抑制效果。此外,若在生產階段中降低培養溫度(28→23oC),則可增產monacolin K,約為2.18倍。 |
英文摘要 | This study is to investigate the effects of environmental factors on monacolin K production by Monascus ruber ATCC 18199. The results of shake-flask experiments showed that the best cultivation occurred while transferring 10﹪spores suspension after 48 h of pre-culture to the production medium. It was found that the pigmentation of the strain used initiated at day 2, followed by monacolin K production at day 6. With an increase of glycerol as 176.4 mg/L, monacolin K production was enhanced to be 2.7-fold of that in the control group. Furthermore, when the ratio of glycerol to peptone (i.e., G/P ratio) was raised up to 33, monacolin K production was maximized as 3.52-fold of that in the control group. However, there was an inhibition if the G/P ratio was 100 or higher, probably due to the excess of nitrogen source. Besides, it was interesting to find a simple temperature-shift method (28-23oC) valuable to the process, where monacolin K production was further increased to be 2.18-fold of that in the control group. |
