中文摘要 | 台灣地區於2009年2月再次爆發豬隻口蹄疫疫性,而引起本次疫情的口蹄疫病毒株(O/TAW/YL/2009),雖仍屬親豬型口蹄疫病毒,但與1997-1998年的口蹄疫病毒株相較,其核酸變異性已達10%。本試驗之目的在於評估已使用超過10年的口蹄疫疫苗株(O/TAW/98)對於2009年新分離株病毒的保護效力。我們使用10頭8週齡無口蹄疫抗體之第二代SPF小豬,先以市售口蹄疫不活化疫苗免疫,再於4週後以口蹄疫新分離株(O/TAW/YL/2009)病毒進行攻毒試驗,並採集豬隻血液、口水及糞便,進行病原及抗體檢測。實驗結果顯示,對照組(未免疫組)豬隻在攻毒後,除出現典型口蹄疫水疱病變及血清口蹄疫病毒非結構蛋白(nonstructure protein; NSP)抗體陽轉外,在其血液、口水及糞便等檢體中均可檢出口蹄疫病毒。另一方面,免疫組豬隻在攻毒後,均未出現臨床症狀,豬隻檢體也未檢出病毒或呈現血清NSP抗體陽轉現象。因此,現行口蹄疫疫苗對2009年新分離株病毒仍具有保護作用。 |
英文摘要 | A sporadic outbreak of foot-and-mouth disease (FMD) occurred in Taiwan in February 2009 and the virus isolated in this outbreak, O/TAW/YL/2009, is a porcinophilic serotype 0 virus similar to those isolated in the 1997 major outbreak. However, the 2009 virus exhibited about 10% nucleotide differences from the FMD virus isolated in 1997-1998. The aim of this study is to evaluate the protective efficacy of currently used vaccine (O/TAW/98) against challenge infection with O/TAW/YL/2009. Ten 8-week-old, SPF pigs that were free of FMD-antibody were vaccinated with the vaccine prepared from O/TAW/98 and then challenged with OITAW/YL/2009 four weeks post vaccination. Samples were collected from the blood, saliva and feces of the pigs after the challenge to detect the antigen and antibody of the virus. The results showed that all unvaccinated pigs showed typical vesicular lesions after the challenge and showed seroconversion to NSP-antibody positive after the challenge. Moreover, FMD virus could be detected in the blood, saliva and feces from the unvaccinated pigs after the challenge. In contrast, all the vaccinated pigs remained healthy after the challenge. No FMD virus could be detected in the vaccinated pigs by RT-PCR after challenge and no seroconversion to NSP-antibody positive was observed. These results demonstrate that the currently used FMD vaccine (O/TAW/98) still confers the pig significant protection against challenge infection with O/TAWIYL/2009. |
