| 中文摘要 | 本研究建立銀葉桉(Eucalyptus cinerea F. Muell. ex Benth.)矮性實生變異株之初代培養。取一年生銀葉桉之單節培殖體,以1%的NaOCl消毒10分鐘後培養可獲得最高的萌芽率。培養基中加入1 g/l聚乙烯比咯酮(Polyvinylpyrrolidone,PVP-40T)抑制褐化效果最好。培殖體以在頂端10cm的腋芽萌芽率最高,並以夏季七月所取的褐化情形最少,污染率最低且萌芽率最高。銀葉桉初代培養基以MS(Murashige and Skoog, 1962)全量、0.1 mg/l BA、0.01 mg/l NAA、1 g/l PVP-40T、蔗糖30 g/l、洋菜8 g/l、pH值5.7的配方最佳。 |
| 英文摘要 | In this experiment, the initial culture of Eucalyptus cinerea ‘Mini’ was established. The sprouting rate of one-node explants from one year old stem which was disinfested by dipping in 1% NaOCl for 10 min. The medium with 1 g/l polyvinylpyrrolidone inhibited the browing of explants more efficiently. The sprouting rate of lateral shoot of one-node cuttings from the upper 10cm of the stem was the highest. In addition, explants harvested in July resulted in the least browning and contamination. Moreover, the best medium was Ms basal medium containing 0.1mg/1 BA, 0.01 mg/1 NAA, 1 g/l PVP-40T, 30 g/l sucrose and 8 g/l agar, and pH at 5.7. |
