中文摘要 | 本研究利用PCR-RFLP (polymerase chain reaction-restriction fragment length polymorphism) 方法分析粒線體DNA的細胞色素b (cytochrome b) 基因,以建立臺灣產鹿科動物肉類製品的鑑定基準,並協助野生動物保育法執行。利用13種限制內切 對14份包括水鹿、梅花鹿、山羌、紅鹿、牛及豬肉樣本進行單切作用,結果依片段長度多型態性的一致性可分成8個單倍基因型(haplotype) ,每個種類都有其特有的單倍基因型,可用來作為種類鑑定之用。根據研究結果,本文提出利用粒線體DNA及PCR-RFLP方法辨識臺灣產鹿科動物的簡單鑑定流程,並探討方法的適用性及優缺點。 |
英文摘要 | We used the restriction fragment length polymorphism analysis (RFLP) of mitochondrial DNA cytochrome b gene amplified by polymerase chain reaction (PCR) to identify meat procucts of cervids in Taiwan. Thirteen restriction enzymes were used to assay the RFLP variation of 14 samples belonging to Formasan sambar deer, Formasan sika deer, Formasan muntjac, red deer, domestic pig and domestic cattle. We detected eight mtDNA haplotypes which are species-specific and useful for their forensic identification. Accordingly, we propose a simple procedure via mtDNA and PCR-RFLP method to identify the species of the meat products of the cervids in Taiwan, and discuss its application, advantages and shortcomings. |
