中文摘要 | 哺乳動物卵巢之濾泡於出生後開始一連串之細胞增生、分化以及生理與生化作用,例如濾泡壁外圍基質組織之重組、濾泡液之形成,以及濾泡細胞表現激性腺素受催/動情素受體等。動物未達發身年齡其濾泡內之卵母細胞休止於第一次減數分裂之前期,染色質散佈於一巨大之原泡核,待動物發身後,卵母細胞與包被在外之卵丘細胞受腦下垂體分泌之FSH和LH刺激而成熟;但僅有少部分之濾泡能發育至成熟階段而排卵,其餘大多數則閉鎖而退化。家畜卵母細胞之成熟,乃指其原泡核崩解(germinal vesicle breakdown, GVBD),自第一次減數分裂前期進行至排出第一極體而達第二次減數分裂中期(metaphase Ⅱ, MⅢ)的階段,過程須經由成熟促進因子(maturation promoting factor, MPF)之活化、細胞骨架、激性腺素、性腺素以及各類生長因子之協同作用來完成。高環境溫度為造成家畜夏季不孕之主要因素之一,因其造成卵母細胞之細胞核、細胞膜、胞器及細胞骨架之改變,此將導致其發育能力降低,而此一問題已成為畜產業亟待解決的難題之一。 |
英文摘要 | The development of ovarian follicles occurs postnatally in most domestic species. It starts from the growth of the primordial follicles and activation of the subsequent development requires a series of cellular, subcellular and molecular transformations. Simultaneously, the enclosed oocyte grows and then arrests at the prophase of first meiosis with large germinal vesicle (GV) insides. However, the maturation of oocytes would not occur until the stimulation of gonadotropins after puberty. Not only nuclear progression but also cytoplamic maturation are required for oocyte capacitiation and/or maturation. The growth and maturation of oocytes and the subsequent development are regulated by the maturation promoting factor (MPG) and the cytoskeleton as well as by GnRh, gonadotropins, sex hormones, cytokines and/or other growth factors. It has been known that environmental factors, such as temperature, affect this complicated process. It has also been suggested that alternations of the nuclei, cytoplasmic membranes, organelles and cytoskeleton by an elevated ambient temperature reduce the reproductive performance of the ammonals. Therefore, improvement of the developmental competence of gametes and embryos during hot seasons becomes a challenge in animal production industry. |
