中文摘要 | 自本省朴子、埔里及霧峰三地分別採得玉米穗蟲(Helicoverpa armigera)、甜菜 夜蛾(Spodoptera exigua)及斜紋夜蛾(S. litura)等三種夜蛾科害蟲之罹病蟲體,經蔗糖梯 度離心純化可得三種核多角體病毒(nucleopolyhedrovirus),進行病毒形態,構造、生化及 生物特性之鑑定與比較。包涵體之形態、構造及每個病毒粒子內所含有之核蛋白鞘數量間差 異頗大。其中以斜紋夜蛾核多角體病毒包涵體最大,平均直徑為1.61±0.32um,而甜菜夜蛾 病毒則為1.07±0.16um,此二病毒粒子內均含多個核蛋白鞘(nucleocapsid)同屬MNPV;另玉 米穗蟲則是最小且為單一核蛋白鞘(SNPV),平均直徑為0.79±0.22um。三種病毒核蛋白鞘平 均大小依序為332.26±13.55 x 47.16±1.42, 297.40±9.14 x 46.72±4.00及319.80± 7.80 x 44.45±4.54um。利用SDS-PAGE之方法分析包涵體蛋白質,得知此三種病毒在微量蛋 白質上有明顯差異,然主要蛋白質之分子量極為接近均在31 kDa左右。利用抗多角體蛋白質 之多源抗體進行酵素聯結免疫吸附檢定法(ELISA),顯示三種病毒之多角體蛋白質間具有極 高之交互免疫反應(cross immune reaction)。三種病毒之基因組核酸,以內核酸限制�t BamH I、Bgl II、EcoR I、Hind III、Pst I及Xho I切割後,進行核酸片段之比對,得知其 間亦具有明顯之差異,玉米穗蟲、甜菜夜蛾及斜紋夜蛾及斜紋夜蛾三種核多角體病毒之核酸 大小依序約為121.7±21.1, 132.4±2.4,及137.8±1.8 kbp。以接種源飲入法餵予此三種害 蟲之四齡初蛻幼蟲,每隻幼蟲接種相同病毒劑量(10�賀IBs/larva),置於25℃生長箱中培養 ,觀察至幼蟲化蛹。由幼蟲積死亡率得知:三種病毒對本寄主害蟲具極高之致病力,玉米穗 蟲核多角體病毒以接種源飲入法測得其對玉米穗蟲一至四齡幼蟲之半致死劑量分別為4,38, 742,4,710 PIBs/larva,且具高度專一性,對斜夜蛾幼蟲不具致死效果,而對甜菜夜蛾幼蟲 則有15%左右之死亡率;而斜紋夜蛾核多角體病毒對另二種幼蟲尚有30%以上之死亡率,甜菜 夜蛾核多角體病毒則可使玉米穗蟲及斜紋夜蛾幼蟲發生13.7-28.4%之死亡率,而此三種病毒 以飼料表面污染法接種三齡小菜蛾,病毒劑量為7.6x10�嵐IBs/mm�攔iet,置於25℃生長箱 中培養,觀察至幼蟲化蛹。結果發現此三種病毒對小菜蛾不具致病力。 |
英文摘要 | Three nucleopolyhedroviruses isolated from the larvae of Helicoverpa armigera, Spodoptera exigua, and S. litura with nucleopolyhedrosis in Taiwan were characterized and compared morphologically, biochemically, immunologically and pathologically. The polyhedra wre all irregular in shape, measuring ca. 0.79±0.22, 1.07±0.16, and 1.61±0.32 um, respectively under electron microscopic observation. The nucleocapsid is enlongated with parallel sides and two straight ends, measuring 319.80±7.80 x 44.45±4.54, 297.40±91.4 x 46.72±4.00, and 332.26± 13.55 x 47.16±1.42 nm, respectively. There were distinct differences between their viral proteins only in minor bands. The molecular weights of purified undegraded polyhedrins were all approximately 31 kDa. Polyhedrins were assayed by ELISA with polyclonal antipolyhedrin antisera specific to each polyhedrin of HearNPV, SpeiNPV and SpltNPV, demonstrating that there were a close relationship between these viruses. Restriction endonuclease profiles of these viral genomes digested with BamH I、 Bgl II、EcoR I、Hind III、Pst I and Xho I restriction endonucleases indicated that these viruses were markedly different from each other, the genome sizes were estimated to be 121.7±2.1, 132.4±2.4, and 137.8±1.8 kbp, respectively. HearNPV isolate is highly specific and pathogenic to H. armigera larvae, its LD�t values were 4,38,742, and 4,710 PIBs/larva to fourth instar, respectively as assayed with an inoculum-imbibing method. These 3 viruses were not pathogenic to Plutella xylostelld. |
