中文摘要 | 本試驗以落花生栽培種臺南十一號十受粉後七天之幼齡子房柄為材料,培養於添加抗壞血酸(ascorbic acid)及酪素水解物(casein hydrolysate)之MS培養基,經120天培養後調查子房柄先端之生長反應、莢果形成及胚發育情形,期能建立有效率之子房柄體外培養技術,作為拯救落花生種間雜交胚之參考。在MS+60g/l蔗糖培養基中,添加ascorbic acid雖然對子房柄之發根及莢果形成率無顯著的提升效果,但對莢果及胚珠發育卻有促進作用,以添加50mg/l ascorbic acid的莢果與胚珠寬度分別可達839mm與2.6mm。 同時添加酪素水解物與ascorbic acid的試驗結果顯示,酪素水解物與ascorbic acid有顯著的交感作用,ascorbic acid促進子房柄發根及莢果形成的效果因酪素水解物添加量不同而異。150 mg/l酪素水解物配合5 0mg/l ascorbic acid之結莢率最高,達81.3%,其莢果與胚珠的平均寬度分別?29.1mm及3.8mm,但含300 mg/l酪素水解物培養基的莢果及胚珠發育則較差。由組織切片的結果顯示,胚珠寬度小於4mm者仍處於未熟胚階段;胚珠寬度大於4mm則已發育為成熟種子,經無菌播種可得正常植株。 |
英文摘要 | In order to establish the in vitro technique of young pges for rescuing the embtryo of interspecific hybrids, the 7-day-old peg tips of Arcachis hypogaea cv. TN11 were used to evaluate the effect of ascorbic acid and casein hydrolyaste on pod formation and embryo development. Although adding ascorbic in the MS+60 g/l sucrose medium could not significantly raise the percentage of rooting and pod set, medium with 50ascorbic acid ascorbic acid has the best promotion for pod enlargement and embryo development. The width median of pod and ovule cultured in the pure MS medium was 7.0 mm and 1.0 mm, respectively. In the medium with ascorbic acid ascorbic acid, the width median of pod and ovule was 8.9 mm and 2.6mm, respectively. The interaction between ascorbic acid and casein hydrolysate was observed in the medium adding both ascorbic acid and casein hydrolyasate. The effect of ascorbic acid on rooting and pod set was different as casein hydrolyaste concentration. Media containing 150ascorbic acid casein hydrolysate were better than 300 mg/l casein hydrloysate. The most favorable medium, 150 mg/l CH and 50ascorbic acid ascorbic acid were added in MS medium, produced 81.3% pod and the width median of pod and ovule was 10.1 mm and 3.8mm, respectively. The ovule more than 4 mm usually developed into mature seeds and the normal plants will be regrenerated. |
