中文摘要 | 生物體在雌激素缺乏的情況下,會造成骨吸收作用遠大於骨形成作用,並伴隨整體骨重塑速率增加而導致骨質淨流失。女性容易獲得骨質疏鬆症以及發生時間皆在停經前後,暗示了雌激素扮演了相當重要的角色。然而,即使經過了約25年的研究,雌激素的缺乏與骨流失之間的關係仍然尚未明瞭。為了想更加釐清這兩者之間的相關性,本研究室以體外骨髓細胞培養系統來研究卵巢切除後不同時期,蝕骨細胞形成能力以及相關細胞激素如腫瘤壞死因子(TNF-a)、介白素-1ß(IL-1ß)以及介白素-6(IL-6)之濃度變化情形。此外,脈衝式電磁場(PEMF)亦被用來探討是否對蝕骨細胞形成有影響以及用以決定相關細胞激素之最佳收取時間。本實驗中以雌性Wistar大白鼠為實驗動物,在經過雙側卵巢切除手術後4、7及14天後,取其長骨內之骨髓細胞做體外培養。經過10天體外培養後,所形成之類蝕骨細胞以抗酒石酸酸性磷酸酵素染色法(TRAP stain)及凹洞形成分析法(pit formation assay)來加以確認並進行個數計數。另外,並利用酵素免疫分析法(ELISA)分析TNF-a、IL-1ß及IL-6之濃度。由實驗結果得知,在卵巢切除4天及7天後,類蝕骨細胞形成能力沒有改變。而在卵巢切除後14天,類蝕骨細胞形成個數卻反而減少;此點發現與其他學者相關研究恰好相異。由本實驗結果發現到卵巢切除後不同時期,類蝕骨細胞形成能力有不同之變化;且細胞激素之結果亦有一致之表現。 |
英文摘要 | Estrogen deficiency leads to increased bone turnover and to a greater increase in bone resorption than bone formation. The predilection for women to acquire osteoporosis and the temporal proximity to menopause implicate estrogen as an important etiologic agent. However, in spite of over 25 years of investigation, the correlation between estrogen deficiency and bone loss is still controversial. To further investigate this connection, the osteoclastogenic capacity and production of associated cytokines such as tumor necrosis factor-a (TNF-a), interleukin 1-ß (IL-1ß), and interleukin 6 (IL-6) of rat bone marrow cells were evaluated at different time points after ovariectomy. In addition, pulsed electromagnetic fields (PEMF) were applied to investigate whether osteoclastogenesis can be affected by electromagnetic fields and to determine the optimal collection time point for cytokines. Female Wistar rats were subjected to bilateral ovariectomy and primary bone marrow cells were harvested at 4, 7, and 14 days after surgery. Recruitment and authentication of osteoclast-like cells were evaluated by reckoning multinuclear, tartrate-resistant-acid phosphatase positive cells on day 10 of culture and by pit formation assay respectively. During the experiments, cytokines like TNF-a, IL-1ß, and IL-6 were assayed by using enzyme-linked immunosorbent assay (ELISA). The osteoclastogenesis of the bone marrow cells remained unchanged 4 and 7 days after ovariectomy. Fourteen days after ovariectomy, however, the number of formed osteoclast-like cells was decreased. These results suggest that the withdrawal of estrogen at different time points induces different changes in osteoclastogenesis in rats, which is consistent with the variations of concentrations of TNF-a, IL-1ß, and IL-6. |
