中文摘要 | 以Paphiopedilum primulinum授粉後110天的綠果莢播種,8星期後可達52.4%的發芽率,授粉後130-150天者則發芽率降至30.8-34.2%。低鹽類濃度1/4-1/8 MS較1/2 MS有利於種子發芽。鹽類組成分以Thomale GD配方與1/4 MS發芽最為理想,分別為58.6%與56.0%。培養基中添加椰子水對種子發芽有顯著的促進效果,馬鈴薯泥與tryptone次之,香蕉泥則有抑制發芽的作用。添加活性碳有利於種子發芽後原球體的發育。液態懸浮培養比固體培養有較高的發芽率,並且發芽速度較快而整齊。 |
英文摘要 | The optimal seed germination of Paph. primulinum in vitro was 110 days after pollination. Seed germinated well in 1/4 MS, 1/8 MS and Thomale GD media. Among the organic supplements tested, coconut milk 100 ml.L¯¹ appeared to be the best for stimulating germination, followed by 20 g.L¯¹ potato homogenate and 2 g.L¯¹ tryptone, while 20 g.L¯¹ banana homogenate was shown to have a negative effect. The additional of activated charcoal seemed to have a little effect on germination percentage, but promoted further development of the protocorms. Suspension culture resulted in a faster, more uniform and higher germination rate, as compared with solid culture. |
