中文摘要 | 內生性有益菌劑對於人畜之效益及其商用價值,已廣為所知。本研究主要目的為篩選適於人、畜用之內生性有益乳酸菌,利用添加 lysozyme, polymycin B, chloram-phenicol 及 rifampicin 之選擇性培養基,可由自然界及商業用菌劑篩選乳酸菌株,再分離生長快速、耐低 pH、耐熱及膽鹽,可產生胞外多醣體及高量乳酸等持性的菌株。藉由選定具有潛力之菌株,進一步經小豬腸道定殖試驗及菌種鑑定,即可做為商品量產之乳酸菌。目前篩選結果,顯示具優良特性的菌株如編號P0002、PLF301、PLF304、PLF502經鑑定為 Lcb. casei ss rhamnosus、Streptococcus mitis 及二株 Enterococcus faecium 等菌株適合添加於豬飼料中。BP70121、BP70162、BP70092、BP70093 等菌株鑑定為 Lcb. bifermentan、Lcb. pentosus、Lcb. plantarum、Lcb. casei ss rhamnosus,可做為供人類食用的菌劑。 |
英文摘要 | The benefits and commercial value of probiotic microorganisms in pharmaceutical preparations for humans and livestock have been well acknowledged. The objective of this study was to select promising probiotic lactic acid bacteria (LAB) strains for human and pig pharmaceutical preparations. Selected media made from several combinations of lysozyme, polymycin B, chloramphenical, and rifampicin were used to isolate promising LAB. Potential strains for industrial application could be determined by the following characteristics: morphology, growth rate, lactic acid productivity, antibiotic resistance, outer cellular polysaccharide and tolerance of temperature, bile salt, and high H□ concentration. The LAB strains selected should be further tested for piglet intestinal colonization before commercial use. The results showed that strains P002, PLF301, PLF304, and PLF502, identified as Lactobacillus casei ss rhamnosus, Streptococcus mitis, and two strains of Enterococcus faecium, respectively, were suitable to add to livestock feed. The other strains with less antibiotic resistance were screened and isolated for possible human usage. The strains demoninated of BP70121, BP70162, BP70092, and BP70093 were Lcb. Bifermentan, Lcb pentosus, Lcb. Plantarum, and Lcb. Casei ss rhamnosus, respectively. |
