中文摘要 | 根管治療失敗有時需藉根尖手術合併逆向充填,才能達到治療病灶保留牙齒的目 的。由於根尖逆充填材料會與根尖組織細胞直接接觸,因此對於材料是否具組織相容性必須 加以仔細探討。 本實驗是用瓊脂覆蓋法探討五種根尖逆充填材料:IRM, Super-EBA,zinc oxide-eugenol,amalgam 及 Cavit 等,對於初代培養之人類牙齦造原纖維細胞( primary human gingival fibroblast )是否具有細胞毒性。結果顯示,經 24 小時之作用後,這些 材料均顯示強弱各不相同的細胞毒性,其中以 zinc oxide-eugenol 最強,Super-EBA 最弱 , 整體而言, 五種測試材料對造原纖維細胞的毒性由強至弱依序排列為: zinc oxide-eugenol > Cavit > IRM > amalgam > Super-EBA,具有統計學上的意義( p < 0.05 )。材料與細胞作用 1 小時後於倒立位相差顯微鏡下,即可觀察到細胞形態已產生變 化,從少數細胞失去紡錘形外觀,至整片細胞剝離,其中以 zinc oxide-eugenol 及 Cavit 所引起的變化最為明顯,IRM,Super-EBA 與 amalgam 較不顯著。 |
英文摘要 | The purpose of this study was to evaluate the cytotoxicity of IRM, Super-EBA, zinc oxide-eugenol, amalgam, and Cavit on primary human gingival fibroblasts using an agar overlay method. Cytotoxicity was quantitatively determined by measuring the zone of cell inhibition around the sample after 24 h exposure of fibroblasts to the tested materials. Statistical analyses using one way ANOVA with Scheffe's test revealed that zinc oxide-eugenol had the hightest cytotoxicity followed by Cavit, IRM, amalgam, and Super-EBA (p<0.05). On the other hand, the powder and liquid components of IRM, Super-EBA, and zinc oxide-eugenol cements also showed considerable cytotoxicity on fibroblasts. And the morphological change of the fibroblasts observed under phase contrast microscope included rounding, detatchment, even lysis could be seen within 1 hour of exposure to the test materials. |
