中文摘要 | 本試驗以青剛櫟(Cyclobalanopsis glauca)之未成熟胚為培植體,置入含1 ppm NAA與0.5 ppm BA的MS培養基中,可誘導出無菌苗。分別以未成熟胚及無菌苗之胚軸、子葉為培植體誘導癒合組織,結果:未成熟胚、胚軸在MS添加1 ppm NAA,子葉在MS添加0.5 ppm 2,4-D之培養基中效果最佳,而三種培植體中,以未成熟胚誘導出的癒合組織量最多。當植物生長調節劑不同。誘導出的癒合組織形態也不同,其中以2,4-D誘導出胚性癒合組織,可作為懸浮培養的材料。 |
英文摘要 | Immature embryos of Cyclobalanopsis glauca were used as explants in this experiment. When immature embryos were transferred into MS medium with 1 ppm NAA and 0.5 ppm BA, the highest germination rate was obtained and in vitro seedling material was established successfully. Callus was induced from immature embryo, hypocotyls and cotyledon of in vitro seedlings. MS medium with 1 ppm NAA was the best medium of plant growth regulators for callus induction in immature embryo and hypocotyls, and MS medium with 0.5ppm 2,4-D the best in cotyledon. The largest amount of callus was induced from immature embryos, among all three explants. Phenotypes of the induced callus varied with plant growth regulators. Embryogenic callus induced by 2,4-D could be used as materials for suspension culture. |
