中文摘要 | 本研究利用 phytase screening medium(PSM)配合平板培養(agar plating)技術, 從不同來源之土壤中分離出40株具植酸 陽性反應之細菌菌株,其中以編號E-22之菌株具 有最高之酵素活性。形態學之研究顯示E-22菌株為格蘭氏陰性桿菌,具有莢膜,大小介於 0.5-1.0x0.8-6.0μm之間。經過一系列生化反應之測定,初步鑑定E-22菌株為Klebsiella屬 之細菌,並且暫定名為Klebsiella sp. FM22。 Klebsiella sp. FM22在液體振盪培養下,其植酸 活性在30小時後達到最高,大部份的 植酸 皆以胞外酵素的狀態存在。酵素基本特性之研究顯示,Klebsiella sp. FM22所產生之 植酸 最適作用溫度為55℃,最適作用pH 6.5,於中性pH範圍酵素之穩定性最佳;在熱穩 定性方面,酵素於100℃下保溫1小時後,仍保有10%左右之活性:A1 、Ba 、Cu 、Zn 等金屬離子及EDTA會抑制植酸 之活性,Mg 及Mn 則可促進植酸 活性。 |
英文摘要 | Forty bacterial strains with phytase activity were isolated from different sources of soil by using phytase screening medium (PSM) and agar plating technique. Strain E-22 was found to have the highest enzyme activity among these bacteria. Morphology study indicated that strain E-22 is a gram-negative, capsule-forming bacterium with cell size in the ranges of 0.5-1.0 x 0.8-6μm. According to the results of a series of biochemical test, strain E-22 was identified as Klebsiella sp. FM22. Time courses study showed that Klebsiella sp. FM22 produces the most active phytase after incubated at 37℃ for 30 hrs in PSM broth medium, and most of the phytase was secreted into the medium. The crude phytase produced by Klebsiella sp. FM22 showed the optimal temperature and pH for the activity at 55℃ and pH 6.5, respectively. This enzyme was stable at neutral pH ranges and maintained 10% of its activity after incubated at 100℃ for 1 hr. The activity of phytase was found to be inhibited by A1 、Ba 、Cu 、Zn ions and EDTA, whereas the enzyme activity was enhanced by the presence of Mg and Mn ion. |
