中文摘要 | 由臺中地區採得發生不規則黃化及壞病徵之嘉德利亞蘭病株,以光學及電子顯微鏡法進行研究觀察,在病組織中並未發現齒舌蘭輪點病毒、喜姆比蘭嵌紋病毒或胡瓜嵌紋病毒,維管束組織的縱切片經紫紅核酸染劑染色後以光學顯微鏡檢查結果,在汰皮組織中可發現染成紫紅色的核內含體,這些內含體的形態有些較密實,有些則呈鬆散狀,核仁常被內含體擠到細胞核邊緣,而核質則遭破壞,有病斑中央部位的切片中,細胞核呈中空而內含體則不明顯,這此顯微構造上的變化與電子顯微鏡鏡檢所得結果相同,在電子顯微鏡的研上,厚切片技術的應用對本病毒之觀察有極大的助益,大小約為120 cm × 45 cm的桿狀病毒顆粒在寄主細胞的胞質或細胞核中發佈或聚集密生,有時整個細胞核充滿病毒顆粒,有些核膜遭破壞而使病毒顆粒發出至細胞質中,在病組織中也有病毒顆粒排列成的輻射狀輪(spoke wheel),由這些核內含體及病毒顆粒的形態特性顯示,這種病毒是發生在嘉德利亞蘭上的一種Rhabdovirus。 |
英文摘要 | A cattleya plant showing irregular chlorotic and necrotic symptoms collected from the Taichung area was diagnosed by light and electron microscopy. In diseased tissue, no odontoglossum ringspot, cymbidium mosaic, or cucumber mosaic viruses were observed. Nuclear inclusion bodies showed purple in infected phloem-related tissues in azure A-stained longitudinal sections of the vascular bundle. Under the light microscope, the aspects of these nuclear inclusion bodies were stacked or lossed, and the nucleolus was crushed to the corner near the nuclear membrane. In some sections, the nuclei were destroyed and appeared hollow. These structural changes caused by viral infection resembled those observed by electron microscopy. The application of semithin-section technology improved the usage of electron microscopy for the observation of this virus. In the ultrastructural study, the bacilliform virus particles, 120 cm × 45 cm in size, were found scattered or aggregated in the cytoplasm or nuclei of infected cells. Some infected nuclei were fulfilled with virus particles, and a few of their membranes was broken and virus particles were released into the cytoplasm. The characteristic spoke wheels were also found in infected cells. According to the results of cytological studies, this virus was supposed to be a rhabdovirus-infected cattelya. |
