中文摘要 | 為了進一步瞭解百合Lilium×Enchantment花粉發育的情形,以液泡期小孢子之花蕾進行活體外之培養,可以獲得發芽率高之成熟花粉;並測試花葯內碳水化合物含量的變化及花粉發芽能力關係。以Heller培養基加上0.01 mg/L NAA,不論是所加蔗糖濃度為1/6M(照光條件下)或是1 M(暗培養條件下),都能在7天內使活體外培養之花粉達到成熟階段(正常活體內花粉需14天達到成熟)。在上述兩種條件下成熟之花粉,其發芽率均在72%左右,而正常活體內成熟之花粉發芽率則為74%。此外,這兩種條件下培養花葯之碳水化合物類型及濃度的變化,與活體內一般生長之花葯相似,顯示本研究已正確的模擬了活體內花粉成熟之條件。反之若培養基內添加光合作用抑制劑DCMU (dichlorodimethylurea),強烈的誘使花葯內可溶性糖分濃度下降,同時也導致了花粉發芽能力大幅降低。進一步證實花粉在發育成熟期間,因為花蕾行光合作用,而提供了花葯發育所需之部分養分。 |
英文摘要 | In order to understand the physiological process of pollen development in Lilium×Enchantment, we observed pollen maturation with flower buds cultivated in vitro, and analyze the anther sugar concentration and pollen germinating ability. The flower buds were cultivated in Heller's medium supplemented with NAA (0.01 mg/l), and pollen maturation achieved within 7 days (although it take usually 14 days in vivo) either under light conditions with 1/6 M sucrose medium or in the dark with 1 M sucrose medium. Pollen germination for the two treatments were; 72±4% and 71±4%, respectively, and were comparable to that of the in vivo matured pollen (74±2%). In both cases, the changes of anther carbohydrate concentrations were similar to that of observed in vivo, indicating that the protocol closely mimics in vivo pollen maturation. Using photosynthesis inhibitor DCMU induced a strong reduction in anther soluble sugars leading to poor pollen germination, confirm that photosynthesis in the flower bud during pollen maturation might partially enhanced anther development. |
