中文摘要 | 在基因的研究上,腫瘤抑制基因(tumor suppressor gene)p�類蘆漪蟔� (mutation)常是導致癌細胞及癌前病灶細胞發生的原因之一。在實驗室如果想要偵 測P�類襤穧]是否有突變,學者多以失去異質性(Loss of Heterozygosity;簡稱LOH) 作為判斷的指標。P�類襤穧]失活化已證實發生於食道癌患者,由於口腔癌和食 道癌的致癌因素頗為類似,相對的基因變異亦可發生於口腔癌。本實驗自18住口 腔癌患者口腔內,以細胞學毛刷(cytology brush)刮除正常粘膜以及口腔癌表面, 收集剝落的上皮細胞,分離萃取genomic DNA,利用聚合脢鏈反應(polymerase chain reaction;簡稱PCR)大量複製P�類襤穧]exon 4與Intron 6位置,以限制脢切離之後, 再經過基因形體限定長度(restriction fragment length polymorphisms;簡稱RFLP)的認 定,比較正常與不正常細胞區基因的異同來作診斷。結果在基因異質性的群體 內,口腔癌細胞基因在exon4位置顯示LOH比率為66(6/9),在intron 6位置顯示LOH 比率為50(3/6)。本研究結果顯示P�奠敦穧]的尖活化與口腔癌的發生是有關的, 且以細胞學毛刷刮取口腔粘膜上皮細胞具有取樣方便且不具侵襲性,應可做為研 究口腔癌診斷的初步篩選。 |
英文摘要 | Allelic detections or/and point mutation in tumor suppressor p�滋� gene are now knownto be associated with the development of cancer in humans. A hallmark of p�滋� is thatboth alleles are generally altered during transformation, which usually represents a lossof heterozygosity (LOH). In this study, 30 healthy dental students and 18 patients withoral squamous cell carcinoma (SCC) were enrolled. Extractions of DNA from oral epithelium of buccal mucosa or cancer surface were sampled using cytologybrush. Two polymorphic restriction sites of exon 4 and intron 6 within p�滋� gene were amplified withpolymerase chain reactions followed by restriction fragment length polymorphism assay.In heterozygous individuals, six of nine (66 ) with oral SCC demonstrated loss of p�滋貪ene heterozygosity at the exon 4 site and three of six (50 )showed LOH at theintron 6 site. These results indicate that inactivation of p" gene is associated withdevelopment and/or progression of oral cancer. . |
