中文摘要 | 台灣地區商業化香菇(Lentinula edodes)菌株種類繁多,如台農1號、中興5號 、中興8號、日本271等品系。為建立台灣地區商業化香菇品系完整之背景資料以供購菌者參考選用,並利於菌種管理,本研究以分子生物技術建立菌種之分子遺傳指紋,試圖找出不同商業化香菇品系間的類緣關係。研究結果顯示,同種的商業化香菇菌株間,可利用逢機擴增多形性DNA (RAPD)分析所得的基因多形性片段圖譜加以區別。本研究一共選用了22條引子進行研究,產生167條肉眼可辨識的DNA片段,並以所得的電泳結果轉換為二元資料,使用NTSYS-PC軟體中的Simqual程序估算遺傳相似係數,再以UPGMA程序建構遺傳相關樹狀圖譜,可將收集到的商業化香菇品系分成下列3群:(1)大陸菌株。(2)台農1號:白殼和紅殼等由國人自行育種成功的品系。(3)日本271、中興5號、中興8號、黑金剛、中溫600等源於日本的品系。利用RAPD技術不但可應用於現有商業化香菇品系間的類緣關係探討,更有助於香菇育種技術的改良,進行新品系的遺傳特性鑑定。 |
英文摘要 | The shiitake, Lentinula edodes (Berk.) Pegler, is the most widely cultivated edible mushroom in Taiwan. Since there are many commercial shiitake isolates in Taiwan, a sensitive and reliable method is needed to differentiate and group these isolates. In this study, the genomic DNA from 25 commercial shiitake isolates was amplified using random amplified polymorphic DNA (RAPD) technique and the polymerase chain reaction (PCR) with single primers. All the amplifications revealed scorable polymorphisms among the isolates, and a total of 167 bands amplified from 22 arbitrary decamer primers were recorded with binary codes. UPGMA (unweighted pair-group method using arithmetic method) cluster analysis was used to divide the isolates of L. edodes into three distinct clusters: (1) No.135 and 939 isolates from China; (2) Tainung No.1 "white cap" and "red cap" isolates; (3) Japanese 271, Jongxing 5, Jongxing 8, Hey-King-Gang and Jong-Wen 600 isolates from Japan. The results indicate that the molecular-genetic markers obtained from the RAPD assay can be used to differentiate commercial shiitake isolates in Taiwan and have potential applications in mushroom breeding and genetic identification of new isolates. |
