中文摘要 | 本研究利用平板培養技術及小滴結冰法由蔬菜葉面分離出五株具有冰核活性之格蘭氏陰性細菌,其中以 INA-3 顯示最高之冰核活性,此菌經初步鑑定為 Pseudomonas sp INA-3,其閾值溫度 (threshold temperature)為-4.8℃。培養條件之研究顯示,當 Pseudomonas sp.INA-3接種於 King's broth 在 22℃ 下培養至對數生長期之末期,細菌培養液具有最高之冰核活性。在 35℃下培養之 Pseudomonas sp.INA-3,其培養液之冰核活性可經由低溫 (10℃)之誘導而提高。添加 Pseudomonas sp.INA-3細胞於含 5-20%蔗糖之水溶液中可提高其冰核溫度,使得在-6℃不易結冰之蔗糖溶液提早結冰。 |
英文摘要 | Five gram-negative bacterial strains with ice nucleation activity were isolated from the leaves of vegetable by using King's agar plating and droplet-freezing assay. Among those bacteria, the strain INA-3 showed the highest ice-nucleation activity and was identified as Pseudomonas sp. INA-3. The threshold temperature of ice nucleation associated with Pseudomonas sp. INA-3 is -4.8 ~C. Cultivation study showed that the optimum conditions for maximum ice nucleation activity were obtained when cells of Pseudomonas sp. INA-3 were incubated in King's broth at 22 ℃ and harvested at the late log phase. Ice nucleation activity of Pseudomonas sp. INA-3 cells grown at 35 ℃ could be induced by incubating the cell cultures at a temperature of 10 ℃. Cells of Pseudomonas sp. INA-3 were incorporated into sucrose solutions to investigate their effects on freezing. The application of INA bacterial ceils caused freezing of sucrose solutions containing 5-20 % sucrose, which did not freeze under the same conditions without INA bacterial cells. |
