中文摘要 | 紅檜是臺灣重要的造林樹種,但其造林木樹幹分叉情形很嚴重,為探討其分叉情形與遺傳之關係,故進行本研究。研究方法主要係使用逢機擴大多形性核酸(RAPD)進行分析。試驗木取自棲蘭、溪頭及臺東三地區之紅檜造林地,分別選取直幹與分叉之造林木各20株,總共以120株樣本進行分析。使用10個逢機引子,共記錄131條DNA片段,其中104條為多形性(79.4%),27條為單形性(20.6%)。 棲蘭地區直幹紅繪之平均遺傳相似為0.875,分叉幹紅檜為0.860;溪頭地區直幹紅檜之平均遺傳相似性為0.829,分叉幹紅檜為0.788;臺東地區直幹紅檜之平均遺傳相似性為0.853,分叉幹紅檜為0.851。群團分析顯示,直幹紅檜與分叉紅檜並不能分為兩群。 棲蘭、溪頭及臺東地區內的平均相似性分別為0.868、0.809及0.852,顯示出臺灣紅檜三個地區族群無分化現象。 |
英文摘要 | The purpose of this project is to know the genetic variation about the trunk character of Chamaecyparis formosensis, which is a very important tree species in plantation but also caused the problem of tree forms. In this study, Random Amplified Polymorphic DNA analysis was performed to determinate genetic variation between straight and folking trees of Chamaecyparis formosensis. Total 120 samples were used in this study, including 20 straight trees and 20 folking trees from each of the three study areas (Chilan, Chitou and Taitung). Ten arbitary sequenced primers were used and 131 DNA fragments were recorded. Among them, 27 fragments showed monomorphism (20.6%) and 104 showed polymorphism (79.4%). Average genetic similarity coefficient for straight and folking trees in Chilan were 0.875 and 0.860, in Chitou 0.829 and 0.788, and in Taitung 0.853 and 0.851 respectively. Colony analyses showed that straight Chamaecyparis formosensis and folking Chamaecyparis fromosensis can't be separated as two different colonies. Average similarity coefficient for Chilan, Chitou and Taitung area were 0.868, 0.809 and 0.852 respectively. These results suggested that Chamaecyparis formosensis in these three areas were not differentiated significantly. |
