| 英文摘要 | Little is known about the fate of the hypertrophic chondrocytes in growth plates and the mechanisms of cell death in cartilage development. We examined the morphological ultrastructure of hypertrophic chondrocytes of growth plates in mice. The process of cell-death of hypertrophic chondrocytes was studied using a high-pressure frozen and freeze-substituted fixation method. Growth plates were obtained from the distal femur in mice. The tissues were frozen in a High Pressure Freezer and freeze-subst tuled, then ultrathin sections were cut and examined by electron microscopy. Hypertrophic chondrocytes appeared very swollen, then the cells ruptured. Electron-lucent nuclei with a small amount of heterochromatin were observed in the cells, and breaks in swollen nuclear envelopes and loss of nuclear matrix were detected. However, In this study, no electron-dense apoptotic bodies were found in the cells. It was found that cell death caused by typical apoptosis does not necessarily occur in terminal hypertrop hic chondrocytes of growth plates in mice. |
