中文摘要 | 本研究以黑白室及舉高型十字迷宮誘發焦慮模式來探討酸棗仁湯及酸棗仁水及95% 乙醇抽取物一次給藥及一週給藥之抗焦慮作用,並以 hexobarbital 誘發睡眠及測定大鼠自 發運動量來探討酸棗仁湯及酸棗仁水及 95% 乙醇一次給藥及一週給藥之鎮靜作用, 最後進 行酸棗仁水抽取物對 APO,HAL,PCPA 及 5-HTP 誘發大鼠自發運動量改變之影響,以探討其 鎮靜作用機轉,並以高效液相層析儀測定大鼠大腦皮質及腦幹中單胺濃度之變化來探討酸棗 仁活性成分抗焦慮作用之機轉。 結果顯示, 在黑白室實驗中,酸棗仁湯及酸棗仁水及 95% 乙醇抽取物一次給藥及一週給藥及酸棗仁總卓撿均可延長小鼠第一次由明室至暗室的時間、 在明室滯留時間及兩室問的穿梭次數,並縮短在暗室的滯留時間。在舉高型十字迷宮實驗中 , 酸棗仁湯及酸棗仁水及 95% 乙醇抽取物一次給藥及一週給藥及酸棗仁總皂鹼均可延長小 鼠在開放臂進入次數及滯留時間的百分比。酸棗仁湯及酸棗仁水抽取物 (0.5,1.0g/kg) 一 次給藥及一週給藥均可延長 hexobarbital 誘發小鼠的睡眠時間及降低大鼠之自發運動量; 酸棗仁湯及酸棗仁 95% 乙醇抽取物一次給藥及一週給藥於較高劑量 (1.0g/kg) 下才具有延 長 hexobarbital 誘發小鼠的睡眠時問及降低大鼠之自發運動量。 APO 及 PCPA 的運動量 興奮作用可被酸棗仁水抽取物 (1.Og/kg) 所抑制;HAL 及 5-HTP 的運動量抑制作用可被酸 棗仁水抽取物 (1.0 g 怕 ) 更抑制。酸棗仁 95% 乙醇抽取物之活性抽出物 (酸棗仁總皂鹼 ) 於 1.Og/kg 下可降低大鼠大腦皮質中 NE,DA 及腦幹中 NE,DA 及 5-HT 的濃度, 並增加 大腦皮質中 VMA,HVA 及腦幹中 VMA,HVA 及 5-HIAA 的濃度。綜合以上結果顯示,酸棗仁湯 及酸棗仁水及 95% 乙醇抽取物一次給藥及一週給藥具有抗焦慮作用, 酸棗仁湯及酸棗仁水 及較高劑量 95% 乙醇抽取物一次給藥及一週給藥具有鎮靜作用, 酸棗仁水抽取物之鎮靜作 用機轉與抑制大腦皮質內倪 techolaminergic system 及增強大腦皮質 serotonergic system 活性有爾。 而酸棗仁總皂鹼之抗焦慮作用機轉可能與降低大腦皮質 NE,DA 及腦幹 NE,DA 及 5-HT 濃度有關。 |
英文摘要 | In this study, we investigated the anxiolytic effect of Suanzaorentang and Suanzaoren after acute and one-week administration by black/white test and elevated plus-maze, the sedative effect of Suanzaorentang and Suanzaoren after acute and one-week administration by hexobarbitalinduced hypnosis and locomotor activity detection. Furthermore, we investigated the sedative mechanism of the water extract of Suanzaozen by combining APO, HAL, PCPA and 5-HTP, and the anxiolytic mechanism of the total glycosides of the ethanol extract of suanzaoren by detecting the changes ofmonoamines and its metabolites in the rats' brain. In the black/white test, the water and 95% ethanol extracts of Suanzaorentang and Suanzaoren after acute and one-week administration and the total glycosides of the ethanol extract of suanzaoren prolonged the first time entry, time spent in the white compartment and the total changes between the two compartments, shorten the time spent in the black compartment. In the elevated plus-maze test, the water and ethanol extracts of Suanzaorentang and Suanzaoaen after acute and one-week administration and the total glycosides of the ethanol extract of suanzaoren prolonged the percentage of entriesand time in the open arms and shorten the percentage of entries and time in the close arms. The water and higher dosage of ethanol extracts of Suanzaorentang and Suanzaoren after acute and one-week administration prolonged the hexobarbital-induced sleeping time in mice and decreased the locomotor activity in the rats. The hypermotilities induced by APO and PCPA were decreased by the water extract of suanzaoren and thehypomotilities induced by HAL and 5-HTP were augumented by the water extract of suanzaoren. The total glycosides of the ethanol extract of suanzaoren decreased the levels of NE and DA in the cortex, the NE, DA and 5-HT in the brain stem, and increased the levels of VMA, HVA in the cortex and the VMA, HVA and 5-HIAA in the brain stem. These results suggested that the water and 95% ethanol extracts of Suanzaorentang and Suanzaoren after acute and one-week administration possessed anxiolytic and sedative effects. The sedative m0echanism of the water extract of suanzaoren was related to the decrease in the catecholaminergic system activity and the increase in the serotonergic system activity. The anxiolytic mechanism of the total glycosides of the ethanol extract of suanzaoren was related to the decrease in the levels of NE, DA in the cortex and NE, DA, 5-HT in the brain stem, and the increase in the levels of VMA, HVA in the cortex and NE.
