英文摘要 | The penetration of sperm into the zona pellucida is a major process of fertilization. Porcine ova were refrigerated in salt solution and the salt-stored ova were washed prior to their usage. Groups of 15 desalted ova were transferred into a droplet of fertilization 'medium and two droplets in a culture dish were prepared. Sperm suspension were added into both droplets, i.e., 30 ova as an assessment unit for each dosage of the testing semen. Droplets in petri dish were then covered with mineral oil and incubated at 3TC for 15 hours. After incubation, loosely binding sperm were washed away from the surface of the zona pellucida and then examined under a fluorescent microscope for the number of penetrated sperm. A total of 39 semen samples consisting of frozen semen from seven Holstein bulls, four Taiwan water buffalo bulls and four Chinese geese, epididymal semen collected from cauda epididymis of a rat, and fresh semen from five New Zealand White rabbits, five Taiwan native chickens, four Tsaiya ducks and nine Muscovy ducks, was assessed for the penetration ability of sperm. Records of 1540 ova with the number of penetrated sperm revealed that the highest number of sperm penetrated into the zona pellucida were 69, 118, 15, 42, 39, 59, 13, and 0, respectively by dairy cattle, water buffalo, rabbit, chicken, tsaiya duck, muscovy duck, goose, and rat. It implied that salt-stored porcine eggs could be used for the assessment of penetration ability of sperm into the zona. Comparison of penetration results of various individuals within each species, either by counting number of ova penetrated or by counting number of sperm penetrated into the zona, showed that the variation among male individuals existed. Rat sperm bound onto the zona pellucida of salted ova but it did not bind onto the zona of a fresh and cumulus-free ova. Therefore, the use of porcine ova for the assessment of fertility of other farm species is feasible. It could be applied to identify the fertile male from stocks in a short time. |
