英文摘要 | Experiments were performed to assess the role of brain angiotensin III (ANG III) in the regulation of blood pressure and body fluid homeostasis in conscious rats with normal and high sodium intake. Male adult Sprague-Dawley rats received intracerebroventricular ANG III infusion (6 μg/hr) for 2 weeks via Alzet miniosmotic pump were housed individually in metabolism cage and allowed to drink either tap water or l% NaCl. Water intake, food intake and urine flow were measured daily and blood pressure was measured by tail-cuff method every other day. The results showed that intracerebroventricular ANG III produced a similar pressor effect (62 mmHg) in both groups of rats. Comparable increases in water intake (1.8-fold) and urine flow (2-fold), and decrease in urine osmolality were also observed in both groups. However, increased sodium and water retention was found only in rats with high sodium intake despite a significant natriuresis. The pressor, dipsogenic and renal responses to central ANG III persisted during ANG III infusion and returned to control levels 2-3 days after termination of heptapeptide infusion. These results indicate that increased central ANG III exerts pressor effect and two diametrically opposed actions to protect body fluid homeostasis. The latter includes a hypervolemic effect mainly via dipsogenesis and probably vasopressin secretion, and a hypovolemic effect via diuresis in normal condition and an additional natriuresis in sodium loaded state. |
