英文摘要 | Apparent Vmax determinations of NAT activity toward these substrates indicated that these 40 colons could be segregated into 3 distinct phenotypes; rapid (22), intermediate (12), and slow (6) acetylators. In both normal and cancerous colon tissues, significantly greater mean apparent Vmax levels were found in rapid than in intermediate and slow (1.2-2.0-fold) (p<0.001) acetylator phenotypes for the two arylamine substrates. Apparent Km determinations indicated that the human colon NAT from a rapid acetylator had a significantly lower affinity for the arylamine substrates (p<0.05) than the intermediate or slow acetylator groups. Comparisons between mean apparent Vmax values and mean apparent Km values for each of the acetylator phenotypes indicated no significant differences between normal and cancerous colon tissues. The N-acetyltransferase activity was inhibited by 5-fluorouracil: at 0.1 mM 5-fluorouracil, activity was reduced 40% and 2.0 mM 5-fluorouracil inhibited activity more than 70%. This report is the first demonstration of acetyl CoA:arylamine N-acetyltransferase activity occurring in cancerous colon tissues in Chinese. |
