英文摘要 | The proline yield of Corynebacterium glutamicum in fermentation media containing 10% NaCl was 3 times higher than that of the control although the growth of bacteria was depressed. The media containing a lower concentration of NaCl (3%) did not show the same effect. The bacteria tamed by 8% NaCl lost the ability to produce proline if the bacteria were switched to regular media to release osmotic stress. This revealed that osmotic pressure caused by the high concentration of the solute could stimulate the biosynthesis of proline. The optimum concentrations of NaCl, nitrogen source (NH �� Cl) and carbon source (glucose) in the medium were 6.90%, 1.40% and 1.76%, respectively, which were determined by RSM (response surface methodology) analysis using a central composite experimental design. Under these conditions, the production of proline was 0.125 ± 0.045 μ mole/mL, and no significant difference at the 5% level was detected by t-test analysis compared with the predicted value (0.123 μ mole/mL). |
