中文摘要 | 飲食中含有的各種生理活性物質與防癌機制應有及密切相關性,而鑑於水產豐富資源的開發利用本研究即以無血清動物細胞培養模式進行評估各種代表性實用貝類之防癌活性成份。本報告為字鮑魚(Haliotis discus)之熱水萃取物中發現含細胞特異性免疫活性物質。鮑魚熱水萃取物能明顯增強人體融合瘤細胞HB4C5的細胞增生與提高抗體產生量。最終250倍純化之活性本體為33 kDa醣蛋白成分,具熱穩定性而在pH 8有最高活性。另外發現細胞經 staurosporine(16nM) 處理後其細胞增生與提高抗體產生活性被阻斷,推測本免疫活性物質可能與細胞膜上特異性受體作用,經蛋白質磷酸激?活化而影響其細胞增生與抗體分泌。 |
英文摘要 | The relationship between nutrition and the genesis and prevention of cancer has become increasingly evident. We report here that a immunostimulating factor was purified from hot water extracted fraction of abalone, Haliotts discus. Serum-free cultured human hybridoma HB4C5 cells were used to investigate the immunological effects of the fraction in vitro. Cell growth as well as antibody production increased significantly in a dose dependent mailer when fractions were added to the cell cultures. The final 250 fold purified immunostimulating factor was confirmed as a N-linked glycoprotein which has M.W. of about 33 kDa, is thermostable and has optimal bioactivity at a pH value of around 8. Staurosporine (16 nM) blocked the effect of antibody production, which implies that the factor which promotes the production of immunoglubulin in cells may be protein kinase-mediated via the specific receptor. |
