英文摘要 | A simple and sensitive high performance liquid chromatographic method was developed for determination of esculetin (6,7-dihydroxycoumarin) and its glucuronide in rat plasma. Optimal response for esculetin was observed with UV at 342 nm. Separation was achieved on a reversed-phase C-18 column. The mobile phase was acetonitrile-water(20:80, v/v, pH2.8-3.0 adjusted by orthophosphoric acid) at a flow rate of 1mL/min. Plasma samples were collected by cardiac puncture 5 min after esculetin (5mg/kg, iv) administration. The concentrations of total (conjugate and free) and free esculetin were 3.57±0.15 and 0.41±0.038μg/mL (n=6), respectively. |
