中文摘要 | 為探討於3。水溫之下不同浸泡時間對九孔受精卵之發育及引發多倍體之影響,以囊胚期胚胎之形態以及胚胎內之染色體數目為指標,以期找出最適合引發三倍體的條件。結果發現,胚胎發育的正常率隨浸泡時間之增加而降低,尤其當浸泡時間超過20分鐘以上,將導致胚胎大量畸形發育。在引發三倍體方面,抑制第一極體處理組的三倍化比率高於抑制第二極體。在浸泡時間方面,以10-20分鐘之效果最佳,但三倍化百分比僅10%-11%。綜合本試驗對胚胎之正常比率及三倍化比率之試驗結果,以抑制第一極體處理10分鐘為最佳引發條件。 |
英文摘要 | The effect of blocking polar bodies (PB1 and PB2) with low water temperature on the development and ploidy of embryos was studied in the small abalone, Haliotis diversicolor supertexta Lischke. A 3°C cold shock treatment of PB1 groups eggs for 5, 10, 20, or 40 min, resulted in 21%, 17%, 7%, and 0% of normal morphologic gastrulas , respectively. The same treatment of PB2 groups eggs produced 12%, 15%, 3%, and 0% of normal morpho-logic gastrulas, respectively. In the control group, 41.8% cells of gastrulas were diploid (n=32), while spontaneous hap-loids (9.0%), triploids (7.5%) and aneuploids (41.7%) were also observed. In treated groups of PB1 eggs, 10.0% of the cells subjected to 10 min of cold shock at 3°C, and 11.0% of the cells in the 20 min treatment were triploid. The embryo survival rates were 17% and 7% for the 10 min and 20 min, treatments respectively cold shock was not effective for inducing triploidy in PB2 eggs. Treating PB1 eggs for 10 min at 3°C was optimal for induc-ing triploidy. |
