中文摘要 | 為明瞭丙醣還原酮(Triose Reductone,TR)與色胺酸(Trp)褐變反應所形成 之褐色物質的性質及構造等;以TR及色胺酸同濃度(莫耳,M)或1:2M混合液於 80~90℃下加熱以調製褐變物質。由其褐變液之濾紙層析顯示兩者之縮合體因加 熱而生成,將上述混合液測其紫外吸收光譜亦顯示產生了縮合體。 將同(0.5)M或1:2M TR與色胺酸在可通人氮氣之燒瓶中加熱以調製該褐變生成物 (縮合物)之結晶,以活性碳脫色並以乙醇再結晶精製之,其收量為33,將此結晶 進行各種試驗,其結果綜合如下: (1)縮合體之融點為178℃。 (2)紫外線吸收光譜之測定:中性液中在310nm,酸性在302nm,鹼性在312~314 nm呈尖狀極大吸收。 (3)溶解液之濾紙層析試驗:各種溶媒均可檢出其有DPI還元性及ninhydrin陽性之 單一斑點。 (4)紅外線圖譜:在3400cm-�薄A2400cm-�薄A,1720cm-�薄A及1650cm-�薊�近,有 最大吸收。 (5)元素分析結果證明其化學式為: (6)Polarogram:pH2.2時,半波電位為+0.19V,波高為51mm。 (7)NMR圖譜:在δ3.24ppm之2質子的spin結合常數J=6.5HZ有二重線峰(A)δ 4.l~4.5ppm有一質子之廣大多重線(B),δ6~6.7ppm有三質子的廣大吸收(c) ,於δ6.75~7.8ppm內δ7.12ppm處呈峰頂,有六質子的多重線裝U(D),δ8.38 ppm有銳單線(峰)(E),δ10.似ppm有一質子的單峰(F)。此圖譜可支持上列 之構造式。綜合上述實驗結果,得知此褐變生成物(結晶)為N-(2-formyl- 2-hydroxyvinyl)-L-Tryptophan'即所謂的Amino reductone。 |
英文摘要 | The Browning reaction product of triose reductone(TR) and Tryptophan wasobtained by heating the reaction mixture, which was prepared first by dissolvingcompletely 0.5M Tryptophan in 0.2N HCI and then equimolar TR at 80-90 ℃ for 1hr through a nitrogen gas. The yield of condensation product was about 33-40. Properties of the condensation product were examined for elementary analysis,melting point, LJV spectrum, paper chromatogram, IR spectrum, NMR spectrumand polarogram.The results were as follows:1. MeltingpointofTR-Trp:178 ℃.2. For paper chromatography, butanol: acetic acid: water and methanoi: water are fairly suitable as solvent.3. UV absorption peak was estimated at 302nm in acid, 310nm in neutral media and at 312-314nm in alkaline medium.4. In IR spectrum, typical peaks were observed at 3400cm-�� (NH), 2400 cm-�� (enolic OH), 1720 cm- �� (CO of carbonic acid) and 1650 cm- ' (CO of alde hyde).5. Polarograms of TR- Trp was estimated at about +0.19 V of half-wave potential, and 51mm of wave height.6. By estimating elementary analysis, it was indicated that the product had the structure of N-(2-formyl-2-hydroxyvinyl)-L-Tryptophan.7. And by detailed investigation on NMR-spectra. chemical structure of TR. |
